HPA、MMP-9、AGEs、RAGE及HSP70在妊娠期高血压患者血清及胎盘中的表达分析 被引量:14

摘要 目的研究妊娠期高血压患者血清和胎盘中乙酰肝素酶(heparanase,HPA)、基质金属蛋白酶-9(Matrix metalloproteinase-9,MMP-9)、晚期糖基化终末产物(advanced glycation end products,AGEs)、糖基化终产物受体(receptor for advanced glycation endproducts,RAGE)及热休克蛋白70(heatshockprotein70,HSP70)的表达意义。方法选取2016年6月至2017年6月确诊的妊娠期高血压患者55例及正常妊娠期孕妇55例作为妊娠期高血压组和妊娠期正常组。提取2组清晨静脉血和胎盘组织样本进行指标检测,HPA、MMP-9指标水平均采用酶联免疫吸附测定法进行检测,AGEs、RAGE指标采用电化学发光法进行检测,HSP70采用免疫透射比浊法进行检测,比较2组血清和胎盘中HPA、MMP-9、AGEs、RAGE及HSP70水平差异。结果妊娠期高血压组患者血清和胎盘中HPA水平显著高于妊娠期正常组(P<0.05),妊娠期高血压组患者血清和胎盘中MMP-9指标水平显著低于妊娠期正常组(P<0.05),妊娠期高血压组患者血清和胎盘中AGEs和RAGE指标水平均高于妊娠期正常组(P<0.05),妊娠期高血压组患者血清HSP70指标水平显著高于妊娠期正常组(P<0.05),妊娠期高血压组患者胎盘HSP70指标水平与妊娠期正常组对象胎盘HSP70指标水平差异无统计学意义(P>0.05)。结论对妊娠期孕妇进行HPA、MMP-9、AGEs、RAGE及HSP70指标检测对妊娠期高血压的诊断具有重要价值意义。 Objective To investigate the expression and clinical significance of heparanase(HPA),Matrix metalloproteinase-9(MMP-9),advanced glycation end products(AGEs),receptor for advanced glycation endproducts(RAGE)and heatshockprotein 70(HSP70)in serum and placenta of patients with gestational hypertension.Methods A total of 55 patients with gestational hypertension who were diagnosed and treated in our hospital from June 2016 to June 2017 were enrolled as gestational hypertension group,and theother 55 normal pregnant women were enrolled as control group.The venous blood and placenta tissue samples were collected in both groups,and the levels of HPA and MMP-9 were detected by enzyme linked immunosorbent assay,and thelevels of AGEs and RAGE were measured by electrochemical luminescence method,moreover,the HSP70 levels were detected by immune transmission turbidimetry,then the results were analyzed and compared between the two groups.Results The levels of HPA in serum and placenta in gestational hypertension group were significantly higher than those in control group(P<0.05),however,the levels of MMP-9 in serum and placenta in gestational hypertension group were significantly lower than those in control group(P<0.05).And the levels of AGEs and RAGE in serum and placenta in gestational hypertension group were significantly higher than those in control group(P<0.05).Moreover the serum levels of HSP70 in gestational hypertension group were significantly higher than those in control group(P<0.05),however,there were no significant differences in the HSP70 levels in placenta tissues between the two groups(P>0.05).Conclusion The detection of HPA,MMP-9,AGEs,RAGE and HSP70 in pregnant women is of great significance for the diagnosis of gestational hypertension.
