摘要
目的:探究热量限制(CR)对糖尿病模型Kkay小鼠体内氧化应激和血脂代谢的影响。方法:将Kkay小鼠随机分为kkay对照组(kkay组,n=8只),热量限制组(CR组,n=8只);另外8只C57小鼠作为常规对照(C57组,n=8只);kkay组与C57组小鼠给予自由饮食,每天记录下小鼠饮食摄入量,CR组饮食摄入量为kkay组小鼠摄入量的60%,干预持续12周,于干预前,干预后第4周、8周及12周测小鼠的血糖和体重;12周后测各组小鼠定超氧化物歧化酶含量(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的含量及血脂代谢的情况。结果:①Kkay小鼠热量限制后,干预CR组血糖随着干预时间的延长,血糖逐渐下降,干预至第8周后,CR组Kkay小鼠血糖显著低干预前水平(P=0.03),同时较kkay组血糖仍显著下降(P<0.001);②正常对照C57组小鼠和kkay组小鼠随着干预时间的增加,体重逐渐增加,而干预后CR组体重较为稳定,并有略微下降,干预至第12周,CR组小鼠体重低于kkay组;③热量限制后,CR组血清胆固醇(TCH)、甘油三酯(TG)及低密度脂蛋白胆固醇(LDL-C)水平显著降低(P<0.05),其血清高密度脂蛋白胆固醇(HDL-C)与kkay组血清差异无统计学意义,仍低于C57组;④kkay组小鼠血清中超氧化物歧化酶(SOD)水平和谷胱甘肽过氧化物酶(GSH-Px)水平显著低于C57组,而给予12周的热量限制后,CR组Kkay小鼠血清SOD和GSH-Px水平显著改善。结论:热量限制可以降低糖尿病模型Kkay小鼠的血糖水平,提高小鼠血清中SOD、GSH-Px水平,改善Kkay小鼠体内的氧化应激和血脂代谢水平。
Objective: To investigate the effects of calorie restriction on oxidative stress and lipid metabolism in diabetic model kkay mice. Methods:Kkay mice were randomly divided into kkay control group (kkay group, n =8), calorie restriction group (CR group, n =8);8 other C57 mice as routine control (C57 group, n =8) Only);kkay group and C57 group mice were given a free diet, and the daily dietary intake of the mice was recorded. The dietary intake of the CR group was 60% of the intake of the kkay group, and the intervention lasted for 12 weeks. The blood glucose and body weight of the mice were measured at 4, 8 and 12 weeks after the intervention and before the intervention;after 12 weeks, the levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and lipid metabolism were measured in each group. Results:①After caloric restriction in kkay mice, the blood glucose of the intervention group was gradually decreased with the prolongation of intervention time. After the intervention to the 8th week, the blood glucose of the kkay mice in the CR group was significantly lower ( P =0.03). Compared with the kkay group, blood glucose was still significantly decreased ( P <0.001).②Normal control C57 group and kkay group mice gradually increased in weight with the intervention time, while the intervention CR group was stable and slightly decreased, at the 12th week of intervention, the mice in the CR group weighed less than the kkay group.③After caloric restriction, the serum levels of TCH, TG and LDL-C in the CR group were significantly lower ( P <0.05). There was no significant difference between the serum HDL-C and the kkay group, which was still lower than that in the C57 group.④Serum SOD levels and GSH-Px levels were significantly lower in the kkay group than in the C57 group, and serum SOD and GSH-Px levels were significantly improved in the kkay mice of the CR group after 12 weeks of caloric restriction. Conclusion:Caloric restriction can reduce the blood glucose level of diabetic model kkay mice, increase the levels of SOD and GSH-Px in serum, and improve the levels of oxidative stress and lipid metabolism in kkay mice.
