基于NEP-SP调控通道分析芩百清肺浓缩丸治疗感染后咳嗽大鼠的量效关系 被引量:6

摘要 目的分析不同剂量芩百清肺浓缩丸治疗感染后咳嗽模型大鼠的量效关系。方法 80只鼠随机分为空白组(予0.9%氯化钠溶液)、模型组(予0.9%氯化钠溶液)、对照组(复方甲氧那明水溶液,相当于成人每次2粒,每日3次)、芩百1组(相当于成人每次3 g,每日2次)、芩百2组(相当于成人每次3 g,每日3次)、芩百3组(相当于成人每次6 g,每日2次)、芩百4组(相当于成人每次6 g,每日3次)、芩百5组(相当于成人每次9 g,每日3次),每组10只。采用烟熏联合LPS鼻腔滴注法及辣椒素雾化吸入诱导模拟革兰阴性菌感染的方法进行造模,造模成功后连续灌胃10 d。观察造模后2 min内、给药后第5天和第10天咳嗽次数。采用ELISA法检测BALF和血清中速激肽P物质(SP)和中性内肽酶(NEP)水平。结果芩百1组血清SP低于对照组,差异有统计学意义(P <0.05)。芩百2组咳嗽次数与对照组比较,差异无统计学意义(P> 0.05),BALF中SP低于对照组,NEP高于对照组,差异有统计学意义(P <0.05)。芩百4组的咳嗽次数和SP均低于对照组,差异有统计学意义(P <0.05)。结论芩百2组剂量能肺内抗炎和有效止咳,芩百4组剂量能明显缩短咳嗽缓解时间,止咳效果优于复方甲氧那明水溶液。芩百清肺浓缩丸治疗感染后咳嗽作用效果存在正相关的量效关系。 Objective: To analyze the dose-effect relationship of different doses of Qinbai Qingfei Concentrated Pills on PIC rats. Methods: 80 rats were randomly divided into the blank group,the model group,the control group,and Qinbai groups(1 to 5),10 rats in each. Literature method was used to establish the model. After success,the rats were given intragastric administration for 10 days. The levels of SP and NEP in BALF and serum were detected by ELISA. Results: The serum SP of Qinbai 1 group was lower than that of the control group,and the difference was statistically significant(P < 0.05). There was no significant difference in cough frequency between Qinbai 2 group and the control group(P > 0.05). SP in BALF was lower and NEP was higher than that in the control group(P < 0.05). The cough frequency and SP of Qinbai 4 group were lower than those of the control group,and the difference was statistically significant(P < 0.05). Conclusion: The dose of Qinbai 2 group can prevent inflammation and cough effectively. The dose of Qinbai 4 group can significantly shorten the cough relief time and relieve cough better than Asme. There is a positive dose-effect relationship between the dose of Qinbai and the above effects.
