摘要
目的研究表皮生长因子(epidermal growth factor,EGF)干预治疗抗NMDA受体脑炎诱导的神经元损伤的保护作用及机制,为抗NMDA受体脑炎提供新的治疗靶点。方法建立抗NMDA受体脑炎诱导的神经元损伤模型,当神经元细胞培养至第8天,将实验组分为1﹑2 d和3 d 3个时相点,每个时相点15个标本,在实验组中加入EGF(10 ng/m L),应用台盼蓝染色法、免疫组织化学和TUNEL细胞凋亡检测等技术,检测EGF对抗NMDA受体脑炎诱导神经元损伤的保护作用以及细胞凋亡基因caspase-3表达水平变化。结果当谷氨酸浓度为100﹑200μmol/L时,细胞凋亡率高[(2.2±1.014)%、(3.3±0.975)%],而细胞存活率并不低[(77.6±5.039)%、(73.4±4.969)%];在诱导神经元凋亡实验中,凋亡后3 d内,3组细胞的神经元细胞存活率相互比较,差异有统计学意义(P<0.05);3组细胞的神经元凋亡率相互比较,差异有统计学意义(P<0.05);3组细胞的caspase-3基因阳性表达率相互比较,差异有统计学意义(P<0.05)。结论EGF对抗NMDA受体脑炎导致的神经元损伤有保护作用,可能是EGF通过促使凋亡基因caspase-3表达减少,抑制神经元凋亡发生,从而对抗NMDA受体脑炎诱导的神经元损伤起到保护作用。
Objective To study the protective effect and mechanism of epidermal growth factor(EGF)intervention on anti-NMDA receptor encephalitis-induced neuronal injury,and provide a new therapeutic target for anti-NMDA receptor encephalitis.Methods A neuronal injury model induced by anti-NMDA receptor encephalitis was established.When the neuron cells were cultured until day 8,the experimental components were divided into three phases,one day,two days and three days,each phase point 15 samples.In the experimental group,EGF(10 ng/mL)was added to the experimental group,and the protective effect of EGF against NMDA receptor encephalitis-induced neuronal injury was detected by trypan blue staining,immunohistochemistry and TUNEL apoptosis detection.And the expression level of the apoptosis gene caspase-3 was changed.Results When the concentration of glutamic acid was 100,200μmol/L,the apoptotic rate was high[(2.2±1.014)%,(3.3±0.975)%],while the cell survival rate was not low[(77.6±5.039)%,(73.4±4.969)%];in the induction of neurons of the apoptosis experiment,the neuronal cell survival rates of the three groups were compared within 3 days after apoptosis,and the difference was statistically significant(P<0.05).The neuronal apoptosis rates of the three groups were compared with each other.the difference was statistically significant(P<0.05).The positive expression rates of caspase-3 gene in the three groups were compared with each other,and the statistical the difference was statistically significant(P<0.05).Conclusion EGF has a protective effect against neuronal damage caused by NMDA receptor encephalitis.It may be that EGF inhibits neuronal apoptosis and playing a protective role in inhibiting the expression of caspase-3 and inhibiting neuronal damage induced by NMDA receptor encephalitis.
