摘要 目的观察长链非编码RNA RP13-753N3. 1在膀胱癌组织和细胞中的表达,探讨RP13-753N3. 1对CDKN1A(cyclin dependent kinase inhibitor 1A)的促进作用及对膀胱癌细胞增殖和侵袭的影响。方法 q PCR检测16对膀胱癌组织和癌旁组织、膀胱癌细胞株和正常膀胱上皮细胞中RP13-753N3. 1的表达。免疫组化检测膀胱癌组织中CDKN1A蛋白的表达。以RP13-753N3. 1表达量最低的膀胱癌细胞株为研究对象,分为阴性对照组(转染阴性质粒)和RP13-753N3. 1组(转染RP13-753N3. 1质粒),q PCR检测RP13-753N3. 1、CDKN1A mRNA的表达,Western blotting检测RP13-753N3. 1对CDKN1A表达的影响,MTT法和Transwell侵袭实验分别检测RP13-753N3. 1对细胞增殖能力和侵袭能力的影响。结果与癌旁组织相比,膀胱癌组织中RP13-753N3. 1低表达(P<0.01)。与人正常膀胱上皮细胞相比,膀胱癌细胞株中RP13-753N3. 1低表达(P<0.05),J82细胞表达量最低(P<0.01)。膀胱癌组织中CDKN1A蛋白的表达明显下调。与阴性对照组相比,RP13-753N3. 1组J82细胞中RP13-753N3. 1表达明显上调(P<0.01),CDKN1A mRNA表达明显上调(P<0. 01)。与阴性对照组相比,RP13-753N3. 1组细胞的增殖能力明显降低(P<0.05),细胞侵袭能力明显下降(P<0.01)。结论 RP13-753N3. 1在膀胱癌组织和细胞株中表达明显下降,过表达RP13-753N3. 1可明显抑制膀胱癌细胞J82的增殖能力和侵袭能力,其分子机制可能是促进CDKN1A基因的表达。 Objective To observe the expression of long-chain non-coding RNA RP13-753N3.1 in bladder cancer tissues and cells,and to explore the effect of RP13-753N3.1 on cyclin dependent kinase inhibitor 1A(CDKN1A)and proliferation and invasion of bladder cancer cells.Methods The qPCR was used to detect the expression of RP13-753N3.1 in bladder cancer tissues and paracancerous tissues,bladder cancer cell lines and normal bladder epithelial cells.Immunohistochemistry was used to detect the expression of CDKN1A protein in bladder cancer tissues.The bladder cancer cell lines with the lowest expression of RP13-753N3.1 were divided into negative control group(transfection with negative plasmid)and RP13-753N3.1 group(transfected with RP13-753N3.1 plasmid).The qPCR was used to detect the expression of RP13-753N3.1 and CDKN1A mRNA.Western blotting was used to detect the effect of RP13-753N3.1 on CDKN1A expression.MTT assay and Transwell invasion assay were used to detect the effect of RP13-753N3.1 on cell proliferation and invasion.Results Compared with adjacent tissues,RP13-753N3.1 was down-regulated in bladder cancer tissues(P<0.01).Compared with normal bladder epithelial cells,RP13-753N3.1 was down-regulated in bladder cancer cell lines(P<0.05),and the expression level in J82 cells was the lowest(P<0.01).The expression of CDKN1A protein in bladder cancer tissues was significantly down-regulated.Compared with negative control group,the expression of RP13-753N3.1 was significantly up-regulated in RP13-753N3.1 group(P<0.01),and the expression of CDKN1A mRNA was up-regulated(P<0.01).Compared with negative control group,the proliferation ability of RP13-753N3.1 cells was significantly decreased in RP13-753N3.1 group(P<0.05),and the cell invasion ability was significantly decreased(P<0.01).Conclusion The expression of RP13-753N3.1 is significantly decreased in bladder cancer tissues and cell lines.Overexpression of RP13-753N3.1 significantly inhibits the proliferation and invasion of bladder cancer cell line J82 by promoting the expression of CDKN1A gene.
