摘要
目的探讨miR-224调控宫颈癌细胞中的表达水平及其对宫颈癌的的增殖、侵袭和迁移作用。方法采用实时定量PCR比较宫颈癌组织及癌旁组织的miR-224基因表达水平。采用脂质体介导转染方法,分别用miR-224模拟剂和抑制剂转染人宫颈癌细胞HCE1,以空白组及阴性转染组为阴性对照组,观察miR-224对宫颈癌细胞增殖、迁移及侵袭能力的影响。Western-blot检测细胞上皮-间质转化(epithelial-mesenchymal transition; EMT)相关因子E-cadherin、N-cadherin和vimentin的表达。结果肿瘤组织miR-224的表达显著高于癌旁组织(P=0. 002)。miR-224模拟剂较阴性对照组能够有效提高HCE1细胞的增殖能力及细胞愈合面积(P<0.05)。miR-224抑制剂组细胞的增殖能力和细胞愈合面积显著低于对照组(P<0.05),miR-224抑制剂组细胞穿膜细胞数明显少于对照组(P<0.05),而miR-224模拟剂组细胞穿膜细胞数明显多于对照组(P<0.05)。miR-224抑制剂组E-cadherin表达显著高于对照组(P<0.05);而N-cadherin和vimentin的蛋白表达显著低于对照组(P<0.05)。但miR-224模拟剂组中E-cadherin的表达显著低于对照组(P<0.05);而N-cadherin和vimentin的蛋白表达显著高对照组(P<0.05)。结论 miR-224在宫颈癌中呈高表达,通过促进EMT来调控宫颈癌细胞的增殖、侵袭和迁移。
Objective To investigate the expression of microRNA-224 in cervical cancer cells and its role in proliferation,invasion and migration of cervical cancer.Methods Real-time quantitative PCR(qRT-PCR)was used to detect the expression of miR-224 gene in cervical cancer tissues and adjacent tissues.The liposome-mediated transfection method was used to transfect human cervical cancer cell line HCE1 with miR-224 mimic and inhibitor,respectively,and the blank and negative transfection were used as controls.And then the effect of miR-224 on proliferation,migration and invasion of cervical cancer cells was observed.Western-blot was used to detect the expression of epithelial-mesenchymal transition(EMT)markers.Results The expression of microRNA-224 in cancer tissues was significantly higher than that in adjacent tissues(P=0.002).The mi-224 mimic effectively improved the proliferation ability and cell healing area of HCE1 cells compared with the negative control(P<0.05).The proliferation ability and healing area of cells in inhibitor group were significantly lower than those in control group(P<0.05).The number of cell penetrating cells in inhibitor group was significantly less than that in control group(P<0.05),while the number of cell penetrating cells in mimic group was significantly more than that in control group(P<0.05).The expression of E-cadherin in inhibitor group was significantly higher than that in control group(P<0.05),while the expression of N-cadherin and vimentin in inhibitor group was significantly lower than that in control group(P<0.05).However,the expression of E-cadherin in mimic group was significantly lower than that in control group(P<0.05),while the expression of N-cadherin and vimentin in mimic group was significantly higher than that in control group(P<0.05).Conclusion The microRNA-224 is highly expressed in cervical cancer,which regulates the proliferation,invasion and migration of cervical cancer cells by promoting EMT.
