摘要 目的通过研究膀胱癌中ILK、E-cadherin及Twist的表达,探讨其在膀胱癌发生、发展中的作用。方法运用免疫组织化学SP法检测ILK、E-cadherin及Twist在62例膀胱癌和25例癌旁正常组织中的表达,并分析其相关性。结果在膀胱癌和癌旁组织中,ILK的表达率分别为54. 8%(34/62)和16. 0%(4/25),两组相比差异具有统计学意义(χ~2=10. 9,P<0.05); E-cadherin的阳性表达分别为32. 3%(20/62)和72. 0%(18/25),两组相比差异具有统计学意义(χ~2=11. 4,P<0.05);Twist的阳性表达分别为64. 5%(40/62)和4. 0%(1/25),两组相比差异具有统计学意义(χ~2=26. 2,P<0.05)。同时,ILK和Twist在高分化膀胱癌中的表达明显低于低分化癌,两组相比差异具有统计学意义(均P<0.05),而E-cadherin在高分化膀胱癌中的表达明显高于低分化癌(P<0.05)。膀胱癌中E-cadherin与ILK和Twist的表达均呈负相关;而Twist与ILK表达呈正相关。结论 ILK、E-cadherin及Twist蛋白在膀胱癌中均有异常表达,并与膀胱癌的分化程度密切相关,ILK、E-cadherin及Twist可能参与膀胱癌的发生、发展。 Objective To investigate the expression of ILK,E-cadherin and Twist in bladder tumor and to explore its significance in carcinogenesis and progression of bladder tumor.Methods Immunohistochemical SP method was used to detect the expression of ILK,E-cadherin and Twist in 62 cases of bladder tumors and 25 cases of normal bladder tissues adjacent to carcinoma.Correlations between ILK,E-cadherin and Twist expression were analyzed.Results The expression rates of ILK in bladder cancer and adjacent tissues were 54.8%(34/62)and 16.0%(4/25),respectively,and there was significant difference between the two groups(χ^2=10.9,P<0.05).The positive expression rate of E-cadherin was 32.3%(20/62)and 72.0%(18/25)in bladder cancer and adjacent tissues,respectively,and there was significant difference between the two groups(χ^2=11.4,P<0.05).The positive expression rate of Twist was 64.5%(40/62)and 4.0%(1/25)in bladder cancer and adjacent tissues,respectively(χ^2=26.2,P<0.05).Meanwhile,the expression of ILK and Twist in highly differentiated bladder cancer was significantly lower than that in poorly differentiated bladder cancer(both P<0.05),while the expression of E-cadherin in highly differentiated bladder cancer was significantly higher than that in poorly differentiated bladder cancer(P<0.05).E-cadherin was negatively correlated with the expression of ILK and Twist in bladder cancer,while Twist was positively correlated with the expression of ILK.Conclusion ILK,E-cadherin and Twist proteins are abnormally expressed in bladder cancer and are closely related to the degree of differentiation of bladder cancer.ILK,E-cadherin and Twist may be involved in the occurrence and development of bladder cancer.
