微小RNA-132对白血病K562细胞增殖及凋亡的影响 被引量:2

摘要 目的研究miRNA-132对白血病细胞株K562增殖、凋亡的影响,探讨其可能的作用机制。方法应用Lipofectami-ne TM 2000转染K562细胞,实时荧光定量PCR检测转染后miRNA-132的表达。CCK-8及流式细胞术检测转染后对K562细胞增殖及凋亡的影响。Western Blot法分别检测SRIT1及P53的表达量变化。结果 miRNA-132在正常细胞中的表达明显低于在K562细胞株中的表达,与空白对照组和阴性对照组相比,转染组miRNA-132的表达量明显升高。转染miRNA-132 mim-ics后,K562细胞增殖能力减弱,凋亡率明显增加。与空白对照组和miRNA-132-NC组相比,miRNA-132高表达后,转染组的SIRT1蛋白表达量明显降低,P53蛋白表达量明显升高。结论高表达的miRNA-132对白血病K562细胞有显著的增殖抑制和凋亡诱导作用,其机制可能与其增强SIRT1、P53的活性有关。 Objective To investigate the effect of miRNA-132 on proliferation and apoptosis of leukemia cell line K562,and explore its possible mechanism.MethodsK562 cells were transfected with LipofectamineTM 2000.The expression of miRNA-132 was detected by real-time quantitative PCR.The effect of transfection on proliferation and apoptosis of K562 cells was detected by CCK-8 and flow cytometry.The protein expression levels of SRIT1 and P53 were detected by Western Blot.ResultsThe expression of miRNA-132 in normal cells was significantly lower than that in K562 cells.Compared with the blank group and the negative control group,the expression of miRNA-132 was significantly increased in the transfected group.K562 cells’proliferated and the apoptosis rate decreased significantly after transfection of miRNA-132 mimics.Compared with the blank control group and miRNA-132-NC group,after the high expression of miRNA-132,the expression of SIRT1 protein in the transfected group was significantly decreased,and the expression of P53 protein was significantly increased.ConclusionHighly expression of miRNA-132 significantly inhibits the proliferation of leukemia K562 cells and induces the apoptosis,which may be related to the enhancement of SIRT1 and P53 activity.
