Simultaneous Determination of 5 Coumarin Components in Angelica Dahurica from Sichuan by UPLC Method

摘要 目的建立UPLC法同时测定不同加工方法和不同产地川白芷中欧前胡素、异欧前胡素、氧化前胡素、水化氧合前胡素、佛手柑内酯等5种香豆素类成分的含量测定方法。方法色谱柱为Agilent Eclipse Plus C18(2.1mm×100 mm,1.8μm),柱温:30℃,流速:0.3 mL/min;检测波长:300 nm,乙腈(A)-0.1%磷酸水溶液(B)梯度洗脱。结果欧前胡素、异欧前胡素、氧化前胡素、水化氧合前胡素、佛手柑内酯等五种对照品线性范围分别为0.005~0.05μg(r=0.9998),0.003~0.03μg(r=0.9999),0.002~0.02μg(r=0.9999),0.002~0.02μg(r=0.9991),0.0008~0.008μg(r=0.9999)。加样回收率在97.00%~101.72%,RSD在0.66%~1.52%。结论该方法具有良好的精密度、稳定性和重复性,可用于川白芷质量控制。 Objective To establish a method for determination of 5 coumarins in the Angelica Dahurica from Sichuan which with different processing methods and areas by ultra-performance liquid chromatography(UPLC). Methods The determination was performed on Agilent Eclipse Plus C18 column(2.1 mm × 100 mm, 1.8 μm) with gradient elution of acetonitrile(A) and 0.1% phosphoric acid aqueous solution(B) at the flow rate of 0.3 mL/min. The column temperature was 30 ℃ and the detection wavelength was at 300 nm. Results Five coumarins of imperatorin, isoimperatorin, oxypeucedanin, oxypeucedanin hydrate and bergapten had good linearitys in the ranges of 0.005~0.05 μg(r=0.999 8), 0.003~0.03 μg(r=0.999 9), 0.002~0.02 μg(r=0.999 9), 0.002~0.02 μg(r=0.999 1) and 0.0008~0.008 μg(r=0.999 9). The average recovery rates were 97.00%~101.72% with RSD of 0.66%~1.52%. Conclusion The method has accurate results, simple operation, good operation, and can be used in the determination of 5 coumarins in Angelica Dahurica from Sichuan effectively.
