摘要
目的通过不同的造模方法,探索制备胃食管反流鼠模型。方法 60只雄性SD大鼠随机分为空白组(CON组)、假手术组(SO组)、稀酸灌注配合夹尾组(GER1组)、浓酸灌注配合夹尾组(GER2组)、幽门半结扎贲门松弛组(GER3组)、幽门半结扎贲门松弛并内损组(GER4组),每组10只。2组灌酸夹尾组进行盐酸(pH=3.0、pH=1.0)灌注及夹尾刺激14 d,2组手术组进行幽门结扎贲门松弛,以及贲门内损手术。通过一般观察、白细胞分类计数、病理学检测判断模型是否成功。结果与CON组比较,GER1组和GER2组体质量和白细胞分类计数差异无统计学意义(P>0.05),且食管胃组织病理学与正常大鼠无异。与SO组比较,GER3组和GER4组在体质量和白细胞分类计数差异有统计学意义(P<0.05),通过组织病理学检测,GER3组和GER4组可见上皮层不同程度增厚以及炎症细胞浸润。且GER4组炎症及损伤表现更符合胃食管反流特点。结论幽门半结扎贲门松弛并贲门内损形成的胃食管反流模型制备方法,术后动物死亡率小,模型质量及病变形成率高。
ObjectiveTo establish a rat model of gastroesophageal reflux model(GERD)by different modeling methods.Methods60 male SD rats were randomly divided into control group,low dose acid combined with tail clipping group,high dose acid combined with tail clipping group,pylorus semiligation and cardiomyotomy group,and pylorus semiligation and cardiomyotomy with internal injury group.And each group had 10 rats.The rats in low and high dose acid combined with tail clipping groups were perfused with hydrochloric acid(pH=3.0,pH=1.0)and performed with tail-clamping stimulation for 14 days.Each model were evaluated by general observation,white blood cell count and histopathological examination.ResultsCompared with the blank group,there was no difference in weight and leucocyte count between the low and high dose acid combined with tail clipping group(P>0.05).Compared with sham-operated group,there were significant differences in body mass and WBC count between pylorus semiligation and cardiomyotomy group,and pylorus semiligation and cardiomyotomy with internal injury group(P<0.05).Histopathological examination showed that the epithelial layer was thickened with different degrees and had been infiltrated with inflammatory cells in the two operation groups.The operation group were more consistent with the characteristics of gastroesophageal reflux.ConclusionThe GERD rat model established by pylorus semiligation and cardiomyotomy with internal injury has higher quality and lesion formation rate,and relatively low mortality rate after operation.
