Optimization of Water Extraction Process for Ruangan Granules by Orthogonal Test

摘要 目的优选软肝颗粒的水提工艺。方法以干膏得率为评价指标,筛选醋鳖甲、醋龟甲先煎工艺的粒度、时间和加水倍数;采用正交试验法优选醋鳖甲、醋龟甲与其余饮片的合煎工艺,以提取液中柚皮苷含量和干膏量为评价指标,考察加水倍数、提取时间、提取次数。结果醋鳖甲、醋龟甲采用最粗粉、先煎0.5 h的干膏得率最优;柚皮苷质量浓度在3.91~500.00μg/mL范围内与峰面积线性关系良好(r=0.9999),平均回收率为95.17%,RSD为0.90%(n=9)。最佳提取工艺为提取3次,加10倍量水,每次提取1.0 h。结论优选的工艺简便、可行,稳定性好,可用于软肝颗粒的提取制备。 Objective To optimize the water extraction process of Ruangan Granules.Methods With the extraction yield as the evaluation index,the granularity,boiling time and water amount of the first decoction process for Carapax Trionycis and Carapax et Plustrum Testudinis were screened.The orthogonal test was used to optimize the combined decoction process for Carapax Trionycis, Carapax et Plustrum Testudinis and other decoction pieces.With the content of naringin and dry extract yield in the extract as the evaluation index, and the water amount, extraction time and extraction times were investigated.Results The Carapax Trionycis and Carapax et Plustrum Testudinis showed the highest extraction yield by decocted first with 0.5 h and coarsest powder.Naringin showed good linear relationship in the range of 3.91-500.00 μg/mL(r = 0.9999),and the average recovery rate was 95.17%, RSD was 0.90%(n = 9).The optimal extraction process was extracting 3 times,adding 10 folds of water,1.0 h each time.Conclusion The optimized process is simple,feasible and stable,which can be used for the extraction and preparation of Ruangan Granules.
