肉制品中大黄鱼源性成分的鉴定

Identification of Larimichthys crocea-derived Materials in Meat Products

摘要 为食品中大黄鱼源性成分的检测提供参考,根据饲料中鱼源性成分定性检测方法(SN/T2867-2011)合成引物(正向引物:5′-TAAGAGGGCCGGTAAAACTC-3′,反向引物:5′-GTGGGGTATCTAATCCCAG-3′),建立基于PCR技术检测鲜肉及加工肉制品中大黄鱼源性成分的检测方法。结果表明:检测大黄鱼成分的引物具有较好的特异性,可扩增出明显的条带,大小为224bp,与大黄鱼的目的基因条带一致,与NCBI中的大黄鱼线粒体DNA全序列(登录号:AP006002.1,Larimichthys crocea mitochondrial DNA,complete genome)具有99%的匹配率;检测灵敏度高,在样品模板DNA浓度为0有.0较01广n的g/适μL用时性也。可扩增出明显的条带;建立的检测方法能准确检测市售多种肉制品中大黄鱼源性成分。 To provide reference for detecting Larimichthys crocea -derived materials in food processing, primers (5′-TAAGAGGGCCGGTAAAACTC-3′, 5′-GTGGGGTATCTAATCCCAG-3′) were designed by ingredient qualitative method. The detection method of L. crocea -derived materials of raw and processed meats was established based on PCR method. The results showed that the techniques based on the primers were specific for species identification and quantification, and had good sensitivity and wide applicability. The 224 bp band was amplified, which were consistent with target gene of L. crocea . The matching rate was 99% with mitochondrial DNA of L. crocea in NCBI. The band was also amplified when the concentration of template DNA was 0.001 ng/μL. So, The biotechnological method based on PCR was appropriate to identify L. crocea -derived materials in raw and processed meats, and had wide applicability.
