摘要
目的探讨脾多肽注射液中的多肽成分及其碰撞诱导解离规律。方法运用三重四极杆质谱对脾多肽注射液中9种多肽进行质谱多反应监测(Multiple Reaction Monitoring,MRM)扫描分析,对多肽EIAQDFKTDL(TD1)进行Q1scan和MS2扫描分析。结果9种多肽均检测到4种可能的多电荷准分子离子峰,[M+2H]2+的碎片离子最多且大部分为小分子碎片离子;在低碰撞能作用下,TD1主要产生b型和y型碎片离子,在最优碰撞能作用下,TD1最强准分子离子峰产生的碎片离子信息丰富,特征碎片离子峰的响应强度大,适合对TD1进行定性与定量分析。本实验所用方法可对脾多肽注射液中的多肽成分进行分析,推测其碰撞诱导解离规律。结论本文为脾多肽的质谱分析提供了一种灵敏度高,重复性好的方法。
Objective Nine kinds of polypeptides in spleen polypeptide injection were scanned by MRM. Methods Adopting Q1 scan and MS2 by triple quadruple mass spectrometry to explore the polypeptides and the regularities of collision induced dissociation in spleen polypeptide injection.Results The results indicated that four possible multi-charge product ions peaks were detected in 9 kinds of polypeptides of spleen polypeptide injection in the MRM model analysis.The amount of [M+2H]2+ were the most and most of them were small molecular fragments.At low collision energy,TD1 mainly produced b- and y-type fragment ions,and the strongest product ion peak of TD1 produced abundant fragment ion information at optimal collision energy.The characteristic fragment ion peak had a strong intensity,which was suitable for qualitative and quantitative analysis of TD1.In conclusion,this method could be used to analyze the polypeptide components and inferred the regularities of collision induced dissociation in spleen polypeptide injection.Conclusion This study provided a high sensitivity and good repeatability mass spectrometry technique for polypeptides in spleen polypeptide injection.
