畲药铜丝藤植物根和根状茎部位对实验性糖尿病小鼠的降血糖作用研究 被引量:2

摘要 目的探讨畲药铜丝藤植物根以及根状茎部位表现出的降血糖作用。方法观察畲药铜丝藤植物根以及根状茎部位提取液对正常小鼠血糖的影响,利用NS针对四氧嘧啶构建糖尿病小鼠模型,对入选小鼠进行分组,分别设为模型组、水提液组[100、200、300 mg/(kg·d)]以及降糖灵组。1次/d的灌胃频率,时间为2周。在第15天,利用葡萄糖氧化酶法对患者的空腹血糖水平进行检测;取构建成功的高血糖小鼠模型,按照随机数字法则分为五组,模型组,药物治疗组(降糖灵组)作为阳性对照,水提液组[100、200、300 mg/(kg·d)],给药3 h后给予2.5 g/kg葡萄糖进行灌胃,在0 h、1 h、2 h及3 h时采用罗氏优越血糖监测仪检测血糖。结果给予正常的小鼠灌胃药物后,各个时间点血糖值和对照组无明显差别,组间比较无统计学意义(P>0.05);用药前,五组血糖对比,差异无统计学意义(P>0.05)。治疗后降糖灵组和提取液组血糖水平均显著下降(P<0.05),且与模型对照组相比,降糖灵组和水提取液(高、中以及低剂量组)差异具有统计学意义(P<0.05);水提液(高、中、低剂量组)主要是在1 h和2 h对血糖升高有抑制作用(P<0.05),且随剂量增多,降糖效果就越明显。结论畲药铜丝藤跟以及根状茎水提液能够显著降血糖且不影响正常小鼠的血糖。 Objective To investigate the hypoglycemic effect of the plant roots and rhizomes of the peony root. Methods The effects of extracts from the roots and rhizomes of the peony roots on the blood glucose of normal mice were observed. The diabetic mice model was constructed by NS against alloxan,and the selected mice were grouped into the model group, the aqueous extract group [100, 200, 300 mg/(kg·d)] and the hypoglycemic group, with the frequency of gavage once a day, and the time was 2 weeks.On the 15th day,the fasting blood glucose level of the patients was detected by glucose oxidase method. The successfully constructed hyperglycemic mouse models were divided into five groups according to the random number rule, including the model group and the drug treatment group (the phenformin group) as the positive control, the water extract group[100, 200, 300 mg/(kg·d)]. The 2.5 g/kg glucose was gavaged at 3 hours after administration. And the blood glucose was measured by Roche superior blood glucose monitor at 0 h, 1 h, 2 h and 3 h. Results After intragastric administration of normal mice, there was no significant difference in the blood glucose levels at each time point compared with that of the control group. There was no significant difference between the two groups (P>0.05). Before the treatment, the blood glucose levels of the five groups were not statistically significant(P>0.05). After treatment, the blood glucose level of the Jiangtangling group and the extract group decreased significantly(P<0.05), and compared with the model control group, the difference between the phenformin group and the water extract(high, medium and low dose groups) was statistically significant(P<0.05). And there was significant difference between the phenformin group, the water extract (high, medium and low dose groups) and the model control group(P<0.05). The water extract (high, medium and low dose groups) mainly inhibited the increase of blood glucose at 1h and 2 h(P< 0.05), and the effect of hypoglycemia was more obvious with increasing dose. Conclusion The water extract of the phyllostachys pubescens plant root and rhizome can significantly reduce blood sugar and does not affect the blood sugar of normal mice.
