子宫内膜异位症恶变组织中RASSF2A启动子甲基化的表达差异性检测及其临床意义

摘要 目的通过运用甲基化特异性聚合酶链反应(MSP)检测子宫内膜异位囊肿(EC)组织、子宫内膜异位相关的卵巢癌(EAOC)中RASSF2A基因启动子甲基化(以下简称甲基化)的情况。探讨该基因启动子甲基化在子宫内膜异位恶变组织中的表达及临床意义。方法选取在宁波市妇女儿童医院进行手术治疗患者的石蜡包埋标本。30份正常子宫内膜组织,30份EC组织,30例卵巢透明细胞癌(OCC)组织,30份卵巢子宫内膜样腺癌(OEA)组织,比较甲基化在不同组织类型患者中的表达差异。显微镜观察筛选20份EAOC样本,比较甲基化与各种临床病理特征的关系。结果 OCC组织中RASSF2A启动子甲基化阳性率明显高于EC和正常子宫内膜组织(均P <0.05),OEA组织中RASSF2A启动子甲基化阳性率明显高于EC组织和正常子宫内膜组织(均P<0.05),EC组织中RASSF2A启动子甲基化阳性率高于正常子宫内膜组织(P<0.05)。EAOC组织中RASSF2A基因启动子甲基化率与患者年龄、临床分期、组织学分级及临床病理类型无关(均P> 0.05);RASSF2A基因启动子甲基化程度与淋巴结转移有关(P <0.05)。结论 RASSF2A基因启动子甲基化与子宫内膜异位症恶变存在联系。 Objective Methylation specific PCR (MSP) was used to investigate the promoter hypermethylation status of RASSF2A gene in tissues of ovarian endometriosis cysts (EC) and endometriosis-associated ovarian carcinoma (EAOC). To explore the expression of RASSF2A gene promoter hypermethylation (RGPM) in endometriotic progression tissues and its clinicopathological significance. Methods The paraffin embedded specimens of patients with EC and EAOC were selected: 30 normal endometrial tissues, 30 EC tissues, 30 ovarian clear cell carcinoma (OCC) tissues, and 30 ovarian endometrioid adenocarcinoma (OEA) tissues, and expression differences of methylation in different tissue types were compared. Twenty cases of EAOC samples were screened by microscope and the relationship between someclinical pathological characteristics were exproed. Results The positive rate of RASSF2A promoter methylation in OCC tissues was significantly higher than that in EC and normal endometrial tissues (bothP<0.05). The positive rate of RASSF2A promoter methylation in OEA tissues was significantly higher than that in EC tissues and normal endometrial tissues (bothP<0.05), The positive rate of RASSF2A promoter methylation in EC tissues was higher than that in normal endometrial tissues (P<0.05). The methylation rate of RASSF2A gene promoter in EAOC tissues was not correlated with age, clinical stage, histological grade and clinical case type (P>0.05). The methylation degree of RASSF2A gene promoter was associated with lymph node metastasis (P<0.05). Conclusions RASSF2A gene promoter in EAOC tissues was not correlated with age, clinical stage, histological grade and clinical case type (P>0.05). The methylation degree of RASSF2A gene promoter was associated with lymph node metastasis (P<0.05). Conclusions RASSF2A genepromoter methylation was associated with the formation of malignant transformation of endometriosis.
