锈毛莓总三萜超声提取工艺的优化及其抗氧化活性研究 被引量:2

摘要 [目的]研究锈毛莓总三萜最佳提取工艺及其抗氧化活性。[方法]在单因素试验基础上,采用Box-Benhnke对影响锈毛莓总三萜提取率的4个因素进行优化分析,并通过对·OH、DPPH·和ABTS^+·的清除能力考察锈毛莓总三萜的体外抗氧化活性。[结果]在超声温度为80℃、料液比为1∶28(g∶mL)、超声时间为70 min、超声功率为400 W、乙醇体积分数为83%时,锈毛莓总三萜提取率为5.06%,与理论值偏差较小。随着样品浓度的增加,其抗氧化能力逐渐增强,其中对DPPH·和ABTS^+·的清除能力较强,半抑制浓度(IC_(50))分别为0.097和0.047 mg/mL;对·OH有较弱的清除能力。[结论]优选工艺适用于锈毛莓总三萜的提取,锈毛莓总三萜具有较好的抗氧化性。 [Objective] The research aimed to study the optimum extraction technology and antioxidant activity of total triterpenes from Rubus reflexus .[Method]Based on single-factor test, four independent factors were selected as affecting factors for Box-Beknken central composite experiment design.·OH,DPPH· and ABTS +· scavenging assays were carried out to study in vitro antioxidant activity of total triterpenes from Rubus reflexus .[Result] The optimal extraction technology was as follow: extraction temperature was 80 ℃, material-liquid ratio was 1︰28(g:mL),extraction time was 70 min, ultrasonic power was 400 W, ethanol concentration was 83%.Under these conditions, the extraction rate of total triterpenes was 5.06%, with a small deviation from the theoretical value. With the increase of the concentration of sample, its antioxidant ability gradually increased. Among them, the scavenging ability of DPPH· and ABTS +·were more obvious, the half inhibiting concentrations (IC 50 ) were 0.097 and 0.047 mg/mL, the scavenging ability of ·OH was weaker.[Conclusion]The method is reasonable for the extraction of total triterpenes from Rubus reflexus , and the total triterpenes has a better anti-oxidation activity.
