摘要
目的从雄激素-雄激素受体通路探析补肾健脾方对糖皮质激素诱导骨骼肌萎缩的保护作用及其分子机制。方法SD大鼠随机分为正常组、模型组、中药组、美雄酮组。除正常组外,其它组腹腔注射地塞米松(1mg/kg. day)进行造模,同时实验组给予中药或美雄酮干预,共14d。实验结束时,测试大鼠有氧运动能力;检测血清睾酮含量; q RT PCR、Westernblot方法检测腓肠肌AR,MyoD,Myogenin,Desmin基因的表达水平。结果与正常组比较,模型组大鼠的有氧运动能力、血清睾酮含量、AR,MyoD,Myogenin,Desmin表达均显著下降,差异有统计学意义(P <0. 05);与模型组比较,中药组与美雄酮组大鼠的有氧运动能力、血清睾酮含量、AR,MyoD,Myogenin,Desmin表达升高,有显著性差异(P <0. 05)。结论补肾健脾方能促进体内雄激素的合成,上调A-AR-MyoD通路相关因子表达,促进肌细胞的增殖与分化,拮抗DEX对骨骼肌的损伤作用。
Objective Study the protective effect and molecular mechanism of Bushen Jian Pi( BSJP) recipe on the dexamethasone induced skeletal muscle atrophy through the Androgen-Androgen receptor pathway. Methods SD rats were randomly divided into four groups: Normal group,Model group,Chinese medicine( BSJP) group and methandienone( Met) group. Except normal group,other 3 groups were intraperitoneally injected with 1 mg/ml dexamethasone( 1 mg/kg,once a day),while the BSJP rats were treated with traditional Chinese medicine,and Met group with methandienone intervention. Aerobic exercise were tested the day before last day. The concentration of testosterone in serum were measured in by ELISA. The expression of AR,MyoD,Myogenin,Desmin were detected by q RT PCR and Western Blotting. Results Compared with the nomal group,model rats’ aerobic capacity and AR,MyoD,Myogenin,Desmin expression was significantly declined. Compared with the model group,the aerobic capacity and t AR,MyoD,Myogenin,Desmin expression was significantly increased in BSJP group and Met group( P < 0. 05). Conclusion Chinese herbs could promote the synthesis of androgen in the body,increase the expression of A-AR-MyoD pathway related factors,promote the proliferation and differentiation of muscle cells,and antagonize the damage effect of DEX on skeletal muscle.
