从抗炎镇痛解热作用初步探究三黄泻心汤复方合煎制备工艺科学内涵 被引量:5

摘要 目的:对比三黄泻心汤处方药材单煎液、分煎合并液、合煎液在抗炎、镇痛、解热方面的药效差异,初步探究三黄泻心汤复方合煎制备工艺的科学内涵。方法:采用二甲苯致小鼠耳肿试验和角叉菜胶致大鼠足肿胀试验,测定大鼠血清中SOD、MDA含量与炎症组织中TNF-α、PGE_2含量对抗炎作用进行考察;采用热板法和醋酸扭体法对各组的镇痛作用进行考察;采用干酵母制热和内毒素致热对各组解热作用进行考察。结果:与正常对照组相比,大黄(P<0.05)、黄芩(P<0.05)、黄连单煎液对二甲苯所致的小鼠耳肿胀抑制效果均不及三黄泻心汤分煎合并液(P<0.01)与合煎液(P<0.01);黄芩、黄连煎液对角叉菜胶所致的大鼠足肿胀抑制作用不显著,大黄煎液、黄连煎液、三黄泻心汤分煎合并液对SOD活性提高作用不显著,此外其余各组对抑制角叉菜胶所致大鼠足肿胀、提高SOD活性、降低TNF-α、PGE_2含量均显著,其中合煎液作用效果尤为突出。在镇痛作用与解热作用的考察中,合煎液与其他组相比均具有更好的镇痛、解热效果趋势。结论:三黄泻心汤合煎液在抗炎、镇痛、解热作用方面要优于药材单煎液与药材分煎合并液,提示三黄泻心汤采用复方合煎的制备工艺具有一定的科学性,在其现代研究与开发中应当重视复方合煎制备工艺。 Objective: The pharmacodynamic differences among separately decocted liquid, separately decocted then combined fluid, and combined decocted decoction in anti-inflammatory, analgesic and antipyretic effect were investigated.The scientific connotation of combined decocting preparation method of Sanhuang Xiexin Decoction was preliminarily studied. Method: Xylene induced mouse ear swelling test and carrageenan induced rats paw swelling test were carried out. The contents of SOD and MDA in serum and content of TNF-α and PGE2 in inflammatory tissue were detected to investigate the anti-inflammatory effect of each group.The hot plate and acetic acid writhing test were carried out to investigate the analgesic effect. The antipyretic effect of each group was researched via dry yeast heat and endotoxin induced heat. Result: Compared with the normal control group,the inhibitory action of separately decocted liquid of Dahuang(P < 0.05), Huangqin(P < 0.05), and Huanglian for xylene induced mice ear swelling was significantly less than the inhibitory effect of separately decocted then combined fluid and combined decocting preparation decoction(P < 0.01). The effect of Huangqin and Huanglian decoction on carrageenan induced rats paw swelling was not significant. The effects of Dahuang, Huanglian and separately decocted then combined fluid were not significant in increasing the activity of SOD. In addition, other groups can significantly inhibit carrageenan induced rat paw edema, improve SOD activity and reduce TNF-α and PGE2 content. The effect of combined decocted decoction was particularly significant. In the investigation of analgesic effect and antipyretic effect, the combined decoction had better effects compared with other groups. Conclusion: Combined decocted decoction has better antiinflammatory, analgesic and antipyretic effect than separately decocted liquid and separately decocted then combined fluid. It suggests that Sanhuang Xiexin Decoction with compound decoction is scientific. In its modern research and development, the preparation process of compound decoction should be paid attention to.
