摘要
目的:探讨肝苏颗粒对结扎胆总管诱导的胆总管阻塞性肝纤维化大鼠的防治作用及可能的作用机制。方法:采用结扎大鼠胆总管建立胆管阻塞性肝纤维化模型,将大鼠随机分为假手术组(n=12)和模型对照组(n=12)、秋水仙碱组(n=12)和肝苏颗粒组(n=12),灌胃8周,取血检测大鼠肝功能,称重肝、脾、胸腺并计算脏器指数,取部分肝组织行HE染色和Masson染色观察肝脏病理组织变化。结果:与正常对照组比较,模型对照组大鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆红素(TBIL)水平明显升高,肝脏系数显著升高,HE染色和Masson染色显示肝组织出现轻至中度的纤维化。与模型对照组比较,秋水仙碱组和肝苏颗粒组血清ALT、AST、TBIL、ALP水平均明显下降,肝脏系数降低,秋水仙碱组的脾脏系数也明显降低(P<0.05或P<0.01)。病理学检查显示秋水仙碱组和肝苏颗粒组肝脏纤维化程度减轻。结论:肝苏颗粒对大鼠胆总管阻塞性肝纤维化有明显的治疗作用,能明显改善肝功能和肝组织病理损伤,机制与增加胆汁分泌排泄,减轻胆汁淤积程度,减轻氧化应激导致的脂质过氧化损害,从而减少肝细胞损伤有关。
Objective: To observe the potential preventive effect and possible mechanism of gansu Granule on hepatic fibrosis rats induced by bile duct ligation. Method: Hepatic fibrosis model rat was induced by bile duct ligation. Rats were randomly divided into 4 groups: model group, colchicine group, Gansu granule group, and sham-operated group in which the rats were give sham operation. After being administered with drugs for 8 weeks, rat blood was taken to measure the liver function of the rats. And the liver, spleen, and thymus were weighed and the visceral index was calculated. The liver tissues were taken for HE staining and Masson staining to observe the pathological changes of the liver. Result: Compared with the control group, the serum levels of alanine aminotransferase(ALT), aspartate amino transferase(AST), alkaline phosphatase(ALP) and total bilirubin(TBIL) were significantly increased in the model group, as well as the liver index. Histopathology showed mild to moderate fibrosis in liver tissue. Compared with the model group, the levels of serum ALT, AST, TBIL and ALP of colchicine group and Gansu granule group were significantly decreased, as well as the liver coefficient. The colchicine group could also reduce the spleen coefficient(P<0.05, or P<0.01).The lesion of the liver fibrosis of colchicine group and liver granules group were reduced compared with the model group. Conclusion: Gansu granule is effective on hepatic fibrosis rats induced by bile duct ligation, which can significantly improve liver function and morphologically reduce hepatic fibrosis. The mechanism was associated with the increase of bile secretion, reduction of the degree of cholestasis, the reduction of the lipid peroxidation caused by oxidative stress, resulting in relieving the damage of liver cells.
