摘要
目的筛选与乙型肝炎病毒(HBV)DNA聚合酶TP区(HBP-TP)相互结合的特异性多肽并研究其对HBV复制水平的影响。方法以大肠杆菌原核表达的HBP-TP重组蛋白为底物,对噬菌体多肽展示文库进行筛选获得特异性噬菌体并测序其多肽区。统计重复率大于10%的多肽序列进行商品化合成。将合成多肽作为药物处理HBV表达细胞后检测HBV复制水平的变化。结果从噬菌体多肽展示文库中筛选出8条靶向HBP-TP重组蛋白的多肽,在细胞水平的检测中发现其中有1条多肽对HBV的复制有明显的抑制作用。结论获得1条对HBV复制水平具有抑制作用的靶向HBP-TP蛋白特异性多肽。
Objective To identify specific peplides binding to hepatitis B virus ( HBV) DNA polymerase TP region ( HBP-TP) and to study their effects on HBV replication. Methods The phage polypeptide display libraiy was screened using the HBP-TP recombinant protein expressed in Escherichia coll as a sul)strate to obtain the specific phage, the polypeptide region of which was then sequenced. Polypeptide sequences with a repetition rate greater than 10% were counted and commercially synthesized. Changes in HBV replication were detected after treatment of HBV-expressing cells with synthetic peptides as drugs. Results Eight peptides targeting HBP-TP recombinant protein were screened out from the phage display library. One of the peptides was found to have a significant inhibitory effect on HBV replication at cellular level. Conclusions A HBP-TP protein-targeting polypeptide with an inhibitory effect on HBV replication was obtained.
