解脲脲原体感染对精子膜蛋白DCXR表达和精子凋亡的影响 被引量:4

摘要 目的探讨解脲脲原体(Uu)感染对男性不育患者精子膜蛋白双羟基/L-木酮糖还原酶(DCXR)表达和精子凋亡的影响。方法参照世界卫生组织(WHO)标准方法对145例精液标本进行精液常规分析。Western blot和免疫荧光分别检测人精子DCXR蛋白的表达和定位,采用TUNEL法检测精子凋亡。对Uu感染阳性者进行敏感抗生素治疗,分析治疗前、后的不育症患者精液各项指标的变化。结果 Uu阳性不育组精子DCXR/β-actin平均光密度和精子DCXR阳性率均低于Uu阴性不育组(P<0.05),治疗后Uu阳性不育组的DCXR/β-actin平均光密度和精子DCXR阳性率均明显高于Uu阳性不育组治疗前,差异有统计学意义(P<0.05)。Uu阳性不育组的精子凋亡率高于Uu阴性不育组,治疗后Uu阳性不育组的精子凋亡率明显低于Uu阳性不育组治疗前,差异有统计学意义(P<0.05)。相关性分析提示精子DCXR蛋白表达量和精子DCXR阳性率分别与精子凋亡率呈负相关性(r=-0.427、-0.562,P<0.01)。结论 Uu感染阳性导致精子DCXR蛋白表达减少、精子DCXR阳性率降低和精子凋亡率升高,Uu感染合理治疗后精子DCXR蛋白表达和精子凋亡有明显改善;精子DCXR蛋白的表达具有保护精子并减少精子凋亡率的作用。 Objective To study the effects of Ureaplasma urealyticum(Uu) on the level of DCXR expression and apoptosis in human spermatozoa. Methods According to semen analysis referred to WHO standard, 145 cases were assigned in infertile group. Western blot was used to measure the level of DCXR expression on spermatozoa. The localization of this protein on human spermatozoa was determined by indirect immunofluorescent staining using anti-DCXR antibody. The sperm apoptosis was assayed with TUNEL. After the treatment of sensitive antibiotics, the changes of indicators before and after treatment for Uu infection were compared. Results The level of DCXR protein expression and the DCXR-positive rate in Uu positive group were lower than those in Uu negative group. After treatment, the level of DCXR protein expression and the DCXR-positive rate were significantly higher than before therapy in Uu positive group(P<0.05). The sperm apoptosis rate in Uu positive group was significantly higher than Uu negative group;after treatment, the sperm apoptosis rate was significantly reduced in Uu positive group(P<0.05). Conclusion Uu infection may affect the level of DCXR protein expression and sperm apoptosis that influence fertility. After effective treatment of Uu infection, the sperm protein DCXR expression and sperm apoptosis would be obviously improved.
