SNHG11 promotion in osteosarcoma cell proliferation by activating wnt pathway

目的探讨长链非编码RNA在骨肉瘤中的作用及机制。方法选取10对骨肉瘤组织标本,利用qRT-PCR方法检测SNHG11在骨肉瘤中的表达水平;采用CCK-8实验检测SNHG11对骨肉瘤细胞增殖能力的影响;通过流式细胞术证明SNHG11对骨肉瘤细胞周期的调控作用;最后,利用western blot筛选SNHG11可以调控的信号通路。结果与癌旁组织相比,SNHG11在骨肉瘤组织中高表达(采用配对样本t检验,n=10,t=3.16,P<0.01);沉默骨肉瘤细胞中SNHG11的表达,细胞增殖能力明显下降[scramble组第1天,(0.69±0.03),第6天(2.11±0.19);siRNA1组,第1天(0.72±0.03),第6天(1.41±0.15);siRNA2组,第1天(0.67±0.07),第6天(1.53±0.28);siRNA-1,P<0.01;siRNA-2,P<0.01],而过表达SNHG11,结果相反[NC组第1天(0.25±0.04),第7天(1.83±0.16),SNHG11组第1天(0.27±0.03),第7天(2.55±0.23),P<0.01];SNHG11可以通过诱导骨肉瘤细胞周期进展促进细胞增殖,并且其可以通过激活Wnt信号通路加速骨肉瘤细胞的增殖。结论 SNHG11在骨肉瘤组织中高表达,其可以通过激活Wnt信号通路促进骨肉瘤细胞的增殖。

Objective To investigate the function and mechanism of SNHG11 in osteosarcoma cells.Methods Ten osteosarcoma tissue specimens were collected.qRT-PCR was used to detect the expression level of SNHG11 in osteosarcoma tissues compared with matched non-tumors.CCK-8 assays were applied to test proliferation function of SNHG11 in osteosarcoma cells.The cell cycle was investigated by flow cytometry.Western blot was used to find the regulatory pathway of SNHG11.Results Compared with adjacent tissues,SNHG11 was highly expressed in osteosarcoma(n=10,t=3.16,P<0.01).Expressions of SNHG11 in osteosarcoma cells were silenced,and the cell proliferation capacity was significantly decreased [ scramble group day 1(0.69±0.03),day 7(2.11±0.19);SiRNA1 group day 1(0.72±0.03),day 7(1.4±0.15);SiRNA2 group day 1(0.67±0.07),day 7(1.53±0.28);SiRNA-1,P<0.01;SiRNA-2,P<0.01 ].Overexpression of SNHG11 showed opposite results [ NC group day 1(0.25±0.04),day 7(1.83±0.16);SNHG11 group day 1(0.27±0.03),day 7(2.55±0.23),P<0.01 ].SNHG11 promoted cell proliferation by inducing the progression of osteosarcoma cell cycle,and it accelerated the proliferation of osteosarcoma cells by activating the WNT signaling pathway.Conclusions SNHG11 high expression in osteosarcoma tissues accelerates osteosarcoma cell growth by activating wnt pathway.
