响应面法优化火麻仁水溶性总蛋白提取工艺 被引量:8

Response Surface Method to Optimize the Extraction Process of Water Soluble Total Protein of Fire Flaxen

摘要 试验通过响应面法确定火麻仁水溶性总蛋白的最佳提取方案,试验过程中分析了三个单因素,分别是提取p H、提取时间和料液比,其中确定了火麻仁的提取时间对火麻仁蛋白提取率影响较大,分析得出火麻仁蛋白提取的最佳p H为11.13,最佳料液比为1︰50.35 (g/mL),最佳提取时间为1.56 h,考虑到实际操作的不便,最终校正为最佳pH11,最佳料液比1︰50 (g/mL),最佳提取时间1.5 h,在此条件下得出的火麻仁的总蛋白含量为34.51%,与预测值相近,同时通过高效液相色谱法分析火麻仁中所含的氨基酸种类,证实火麻仁中有多种对人体有益氨基酸,其中精氨酸与谷氨酸含量较高,火麻仁蛋白冻干粉中精氨酸含量约为7.35%。 In this experiment, the response surface methodology was used to determine the optimal extraction protocol for water soluble total protein of Hemp kernel. Three single factors were analyzed during the experiment which were extraction p H, extraction time, and ratio of solid to liquid. Among them, hemp kernel was determined. The extraction time had a great influence on the extraction rate of the protein in the hemp seed protein. The optimal pH was 11.13 for the extraction of the hemp protein, the optimal material to liquid ratio was 1︰50.35(g/mL) and the optimal extraction time was 1.56 h. The actual inconvenience, the final correction was the optimal pH 11, the best material to liquid ratio of 1︰50(g/mL), and the best extraction time 1.5 h, the total protein content of the hemp seed obtained under the conditions was 34.51%, similar to the predicted value, and analysis of amino acids contained in hemp kernels by high performance liquid chromatography confirms that there were many kinds of beneficial amino acids for humans in hemp kernels, in which the content of arginine and glutamic acid was high. The content of arginine in the freeze-dried powder of hemp protein was about 7.35%.
