初诊慢性粒细胞白血病疾病各阶段WT1基因表达量的研究

Study on Expression of WT1 Gene in Different Stages of Newly Diagnosed CML

目的探讨WT1基因在初诊慢性粒细胞白血病(CML)患者疾病各阶段中的表达量的特点.方法通过Taqman荧光探针RQ-PCR方法分别对188例初诊CML的患者的WT1目的基因及ABL内参基因进行检测,利用标准曲线法对WT1基因的表达水平进行绝对定量,检测结果用WT1与10000倍ABL基因拷贝数比值(WT1/10000 ABL)表示.结果初诊188例CML患者中有慢性期158例,WT1表达量范围在4~470 copies/10000 ABL copies;加速期患者26例WT1表达量范围在386~14439 copies/10000 ABL copies;急淋变期患者4例WT1表达量范围在26~667 copies/10000 ABL copies.结论WT1基因在CML患者各疾病期均有表达,并且加速期患者的WT1表达量最高,明显高于慢性期和急淋变期;WT1表达量的多少可以预示CML所处疾病阶段.

Objective To explore the characteristics of the expression of WT1 gene in newly diagnosed chronic myeloid leukemia(CML)patients at different stages of disease.Methods Taqman Fluorescent Probe RQ-PCR was used to detect the WT1 target gene and ABL reference gene in 188 newly diagnosed CML patients.The expression level of WT1 gene was quantified by standard curve method.The results were expressed by WT1 and ABL copy number ratio(WT1/10 000 ABL).Results Among 188 newly diagnosed CML patients,158 were in chronic stage,the expression of WT1 ranged from 4 to 470 copies/10 000 ABL copies,26 in accelerated stage from 386 to 14 439 copies/10 000 ABL copies,and 4 in acute lymphoblastic stage from 26 to 667 copies/10 000 ABL copies.Conclusion WT1 gene was expressed in all disease stages of CML patients,and the expression of WT1 was the highest in accelerated stage patients,which was significantly higher than that in chronic stage and acute lymphoblastic stage;the expression of WT1 could predict the disease stage of CML.
