虾青素对异丙肾上腺素诱导的大鼠心肌纤维化的影响 被引量:3

摘要 目的探讨虾青素对异丙肾上腺素(ISO)诱导的大鼠心肌纤维化的影响.方法SD大鼠腹腔注射异丙肾上腺素(5mg·kg^-1·d^-1),连续注射14d,建立心肌纤维化模型,从建模第2天开始给予虾青素(5mg·kg^-1·d^-1和10mg·kg^-1·d^-1)灌胃,连续21d.实验结束后超声心动图检测大鼠心脏功能,计算心脏指数,Masson染色观察心肌纤维化水平,Western blotting方法检测心肌胶原Ⅲ和转化生长因子-β1 (TGF-β1)蛋白的表达.结果与正常对照组比较,模型组左心室射血分数[LVEF(65.10±3.00)%比(86.54±1.89)%]和左心室短轴缩短分数LVFS[(31.29±2.07)%比(50.66±2.49)%]明显降低(P<0.05),心脏指数[(3.82±0.13)比(3.02±0.08)]升高,心肌纤维化明显,胶原蛋白Ⅲ[(0.58±0.02)比(0.07±0.02)]及TGF-βl[(0.50±0.02)比(0.07±0.02)]表达水平升高(P<0.05);与模型组比较,低剂量虾青素组和高剂量虾青素组LVEF[(71.55±1.27)%比(76.67±2.26)%比(65.1 0±3.00)%]和LVFS[(36.63±0.50)%比(40.35±2.01)%比(31.29±2.07)%]明显升高(P<0.05),心脏指数减低[(3.6 1±0.05)比(3.36±0.06)比(3.82±0.13)](P<0.05),心肌纤维化程度减低,心肌胶原蛋白Ⅲ[(0.41±0.02)比(0.28±0.02)比(0.58±0.02)]、TGF-β1[(0.34±0.02)比(0.14±0.01)比(0.50±0.02)]表达水平明显降低(P<0.05).结论虾青素能够降低异丙肾上腺素诱导的心肌纤维化水平,改善心功能. Objective To investigate the effects of astaxanthin on myocardial fibrosis induced by isoproterenol (Iso) in rats. Methods Myocardial fibrosis model of rats was built by intraperitoneal injection of ISO for 14 days(5mg·kg^-1·d^-1). From the second day on. rats were treated with astaxanthin(5mg·kg^-1·d^-1. 10mg·kg^-1·d^-1) by gavage for 21 days. On the day 23. echocardiography was performed to investigate cardiac fbnction of rats, then rats were sacrificed and the cardiac index was calculated. The myocardial fibrosis degree was observed by Masson staining and the expression of collagen Ⅲ and TGF-β1 were detected by western blotting. Results Compared with the nonnal control group, the myocardial indexes [(3.82±0.l3) vs.(3.02± 0.08 )];the level of cardiac fibrosis increased;the expression of collagen Ⅲ[(0.58±0.02) vs.(0.07±0.02)]and TGF-β1[(0.50±0.02) vs.(0.07±0.02)] were markedly increased in Iso group;whereas the LVEF [(65.10±3.00)% vs.(86.54±1.89)% and LVFS ( 31.29±2.07)% vs.(50.66±2.49)% decreased in Iso group. Treatment with astaxanthin significantly reduced cardiac index.(3.61 ±0.05 ) vs.(3.36±0.06) vs.(3.82±0.13 )];the level of cardiac fibrosis, the expression of collagen Ⅲ[(0.41 ±0.02) vs.(0.28±0.02) vs.(0.58±0.02)] and TGF-β1 [(0.34±0.02)vs.(0.14±0.01) vs.(0.50±0.02)];LVEF[(71.55±1.27)% vs.(76.67±2.26)% vs.(65.10±3.00)%] and LVFS [( 36.63±0.50)% vs.(40.35±2.01 )% vs.(31.29±2.07)% increased. Conclusion Astaxanthin can alleviate myocardial fibrosis and improve cardiac function induced by Iso in rats.
