阿那其根醇提取物对变异性咳嗽患者淋巴细胞增殖作用及炎症因子的影响

Effects of alcohol extract of Anacyclus pyrethrum root on lymphocyte proliferation and inflammatory factors in patients with cough variant asthma

目的探讨阿那其根醇提取物(EEAP)对变异性咳嗽(CVA)患者淋巴细胞增殖及炎症因子的影响。方法使用淋巴细胞分离液获取CVA患者淋巴细胞,在培养液中加入植物凝集素(终浓度为100μg·mL^(-1))以及1、10、100 mg·mL^(-1)的EEAP,经过不同时间培养,采用CCK8法检测淋巴细胞的增殖,ELISA法检测不同炎症因子的水平。结果 EEAP可以促进淋巴细胞的增殖;同时能降低CVA患者血清中炎症因子IL^(-1)3、IL-5、IFN-γ、TNF-α及GM-CSF的浓度,并可升高IFN-γ的浓度(P <0.05)。结论 EEAP可增强患者淋巴细胞的免疫调节活性,同时可调节由Th1细胞分泌的IFN-γ和Th2细胞分泌的IL^(-1)3、IL-5因子的含量水平,有助于提高患者免疫功能。

Objective To determine the effect of alcohol extract of Anacyclus pyrethrum root on lymphocyte proliferation and inflammatory factors in patients with cough variant asthma(CVA). Methods Lymphocytes of CVA patients were obtained with lymphocyte isolation medium. Plant lectin(final concentration at 100 μg·m L^-1)and anachitine alcohol extract(1, 10, and 100 mg·mL^-1) was added to the culture medium. After being cultured for different periods of time, CCK8 method was used to determine the proliferation of lymphocytes.ELISA was used to detect the levels of different inflammatory factors. Results The alcohol extract of Anacyclus pyrethrum root promoted lymphocyte proliferation, decreased the concentration of IL-13, IL-5, IFN-γ, TNF-αand GM-CSF in the serum of CVA patients, and increased the concentration of IFN-γ(P<0.05). Conclusion The alcohol extract of Anacyclus pyrethrum root can enhance the immunomodulatory activity of lymphocytes and regulate the content of IFN-γ secreted by Th1 cells and the level of IL-13 and IL-5 factors secreted by Th2 cells, which is helpful to improve the immunity of patients.
