摘要
目的探讨电针足三里对肠易激综合征腹泻型(IBS-D)大鼠模型肠道炎性物质的影响。方法三月龄SPF级雌性SD大鼠80只,采用随机数字表法分为4组:对照组、模型组、针刺组、假针刺组,每组20只。以高乳糖饲料喂养与束缚应激相结合的方法制备IBS-D大鼠模型,针刺组采用电针足三里治疗,假针刺组选取足三里穴外侧5 mm区域的非穴位区作为针刺点。观察大鼠日常饮食及排便情况,HE染色观察结肠组织病理结构,ELISA定量检测肠道炎性因子IL-1β、IL-6的含量,Western blot检测结肠组织蛋白NF-κB的表达。结果与对照组比较,模型组大鼠2周时体重增长率降低(P<0.05),近端结肠肠道炎性因子IL-1β、IL-6含量、结肠组织蛋白NF-κB表达均增高(P<0.05);与模型组比较,针刺组体重增长率明显上升(P<0.05),近端结肠肠道炎性因子IL-1β、IL-6含量、结肠组织蛋白NF-κB表达均降低(P<0.05);假针刺组与模型组比较差异无统计学意义(P>0.05)。结论电针可以有效改善IBS-D体重增长率,降低结肠黏膜组织炎性因子含量;同时调节结肠组织NF-κB蛋白的表达,减轻炎性损伤。
Objective To investigate the effect of electro-acupuncture at Zusanli(ST36) on intestinal inflammatory substances by using diarrhea-predominant irritable boewl syndrome(IBS-D) rat model. Methods Eighty three-month-old female Sprague-Dawley(SD) rats were randomly divided into four groups with random digital method,i.e.. control group, model group, real acupuncture(RA) group and sham-acupuncture(SA) group, 20 rats in each group. IBS-D rat model was prepared by the combination of high lactose feed and restraint stress. Rats in RA group were acupunctured at Zusanli(ST36) while rats in SA group with the non-acupoint area 5 mm away from Zusanli. The daily diet and defecation of rats were observed. The pathological structure of colon tissue was observed by HE staining. The levels of IL-1β and IL-6 in intestinal tract were detected by enzyme-linked immunosorbent assay(ELISA). The expression of NF-κB in colonic tissue was detected by Western blot. Results Compared with the control group, the growth rate of the model group was significantly decreased at 2 weeks(P<0.05), the levels of inflammatory factors IL-1β, IL-6 and the expression of NF-κB in the colon were significantly increased(P<0.05). Compared with the model group, the body weight growth rate of the RA group increased significantly(P<0.05), the expression of IL-1β, IL-6 and NF-κB in the proximal colon was significantly decreased(P<0.05). There was no significant difference between SA group and model group(P>0.05). Conclusion Electro-acupuncture can effectively improve the growth rate of IBS-D rats and reduce intestinal mucositis, reduce the content of inflammatory factors in colonic mucosa and simultaneously regulate the expression of NF-κB protein in colonic tissue and reduce inflammatory damage.
