复方辛夷滴鼻液对变应性鼻炎大鼠鼻腔灌洗液中细胞因子IL-4、IL-13、INF-γ、TSLP含量及鼻黏膜TSLP影响 被引量:7

摘要 目的:研究复方辛夷滴鼻液对变应性鼻炎大鼠鼻腔灌洗液中细胞因子IL-4、IL-13、INF-γ、TSLP的含量及鼻黏膜TSLP的影响。方法:卵清蛋白(致敏原)+氢氧化铝(佐剂),制成OVA混悬液。将60只大鼠随机分为AR组(A组)、AR+复方辛夷滴鼻液治疗组(B组)、健康空白对照组(C组),每组20只。于基础致敏和局部激发后,观察大鼠行为,记录症状体征积分,并留取各组大鼠鼻腔灌洗液和鼻黏膜,检测鼻腔灌洗液中IL-4、IL-13、IFN-γ、TSLP等的含量及鼻黏膜TSLP的含量。结果:C组大鼠鼻腔灌洗液中IFN-γ相对表达量最低;A组和B组的IFN-γ表达量明显升高,A组较B组相对表达量降低。C组大鼠鼻腔灌洗液中IL-4、IL-13及TSLP的相对表达量最低;A组和B组大鼠的IL-4、IL-13及TSLP表达量明显升高,而A组较B组相对表达量明显升高。各个指标中,B组与A、C两组比较,P<0.05,差异有统计学意义。C组鼻黏膜中TSLP的浸润很少,A组鼻黏膜的TSLP浸润最多,B组较A组TSLP浸润明显减少,但与C组相比,TSLP浸润数目偏高。结论:复方辛夷滴鼻液能够促进AR大鼠抗炎因子IFN-γ的高表达;抑制AR大鼠细胞因子IL-4、IL-13及TSLP的分泌;减少AR大鼠鼻黏膜中TSLP细胞因子的浸润;这有可能是复方辛夷滴鼻液治疗AR的作用机理之一。 Objective:To study the effects of Compound Xingyi Nasal Drops on cytokine IL-4,IL-13,IFN-γ,TSLP and nasal mucosal TSLP in nasal lavage in rats with variable rhinitis. Methods:Egg albumin(allergens)+ aluminium hydroxide(adjuvant)was made into OVA suspension. The 60 rats were randomly divided into AR group(group A),AR+Compound Xinyi Nasal Drops treatment group(group B),health blank control group(group C),20 in each group. After the basic sensitizing and local excitation,the behavior of rats was observed,the symptoms and signs were recorded,and the nasal lavage fluid and nasal mucosa of rats were taken,and the contents of IL-4,IL-13,IFN-γ,TSLP and so on were detected in nasal lavage fluid and the content of nasal mucosa TSLP. Results:The relative expression of IFN-γin nasal lavage fluid in group C rats was the lowest. The expression of IFN-γin group A and group B increased significantly,and the relative expression of group A was lower than that of group B. The relative expression of IL-4,IL-13 and TSLP in nasal lavage of rats in group C were the lowest,and the expression of IL-4,IL-13 and TSLP in group A and group B rats increased obviously,while the relative expression of group A were significantly higher than that in group B. In each index,group B compared with group A and C two groups,P<0.05,the difference was statistically significant. The infiltration of TSLP in nasal mucosa of group C was rare,the TSLP of nasal mucosa in group A was the most infiltrated,and the infiltration of group B was significantly lower than that of Group A,but the number of TSLP infiltration was higher than that of group C. Conclusion:Compound Xingyi Nasal Drops can promote the high expression of anti-inflammatory factor IFN-γ in AR rats;inhibit the secretion of cytokines IL-4,IL-13 and TSLP in AR rats;reduce the infiltration of TSLP cytokines in nasal mucosa of AR rats This may be one of the mechanisms of action of Compound Xingyi Nasal Drops in the treatment of AR.
