摘要 在多细胞生物个体中,细胞增殖和死亡之间的动态平衡是维持生物体内环境稳态的必要条件,也是生物体生长发育的重要条件之一。当体内细胞过度增殖或细胞正常死亡受到抑制时,恶性肿瘤的发生率大大增加。因此,有研究者认为,恶性肿瘤的两大显著特征为细胞无限增殖和死亡抑制[1]。在传统意义上,人们将细胞的死亡分为调控型和非调控型两类。然而,近年来研究发现,细胞坏死也可以像细胞凋亡一样受到遗传物质的调控,这是一种新型的细胞死亡机制。因其由基因控制但细胞形态表现为细胞坏死,所以又称为坏死性凋亡,并且其调控过程不经含半胱氨酸的天冬氨酸蛋白水解酶(caspase)介导。大量的研究证明,坏死性凋亡或程序性坏死在恶性肿瘤的发生、发展、侵袭、转移以及耐药中发挥着重要的作用。本文将对坏死性凋亡机制及其有别于凋亡、坏死等的特点,在肿瘤治疗中的潜在应用等做简单阐述,以期为肿瘤治疗提供新靶点、新思路。
