摘要
为分析中国美利奴羊(新疆型)PTPN11基因的多态性及与细毛羊重要经济性状的相关性,寻找可用于中国美利奴羊(新疆型)选育的分子遗传标记。本研究运用PCR-SSCP技术检测PTPN11基因的多态性,用SAS 8.1软件中的最小二乘方法分析PTPN11基因与羊毛性状的关联性。结果表明,PTPN11基因共检测到了AA、AB和BB 3种基因型,其基因型频率分别为0.28、0.52和0.20;A、B等位基因频率分别为0.54和0.46,A等位基因占优势。χ~2检验显示,PTPN11基因处于哈代-温伯格平衡状态(P>0.05),PTPN11基因位点的多态信息含量为0.37,PTPN11基因在该群体中处于中等程度的遗传变异。不同基因型的个体对弯曲评分有显著影响(P<0.05),对其他性状均无显著影响(P>0.05)。
The aim of this study is to analyze the polymorphism of the Chinese Merino sheep (Xinjiang type) PTPN11 gene and its correlation with the important economic traits of the fine wool sheep,and to find the molecular genetic markers that can be used for the breeding of Chinese Merino sheep (Xinjiang type).In this study, PCR-SSCP was used to detect the polymorphism of PTPN11 gene,and the least squares method in SAS8.1 software was used to analyze the correlation between PTPN11 gene and wool traits. The results showed that the three genotypes of PTPN11 gene were detected as AA,AB and BB,and their genotype frequencies were 0.28,0.52 and 0.20,respectively. The allele frequencies of A and B were 0.54 and 0.46,respectively,A allele was the dominant allele.χ^2 test showed that PTPN11 gene loci was in Hardy Weinberg equilibrium state (P >0.05). The polymorphism information content of the PTPN11 gene was 0.37, therefore, the genetic variation of the Chinese Merino (Xinjiang type) sheep population was in the medium level (P <0.01). Individuals with different genotypes had significant effects on bending score (P <0.05),but had no significant effect on other traits (P >0.05).
