摘要
目的研究miR-214通过调节凋亡蛋白Caspase 3表达而对肝纤维化小鼠的影响。方法将6周龄C57BL/6小鼠随机分为两组:其中实验组注射四氯化碳诱导肝纤维化,对照组注射橄榄油;采用尾静脉注射方法,对小鼠进行miR-214agomir干预;运用H&E和天狼猩红染色观察纤维化模型病理变化;采用Western blot检测实验组和对照组α-SMA和Caspase 3在肝脏的表达水平;实时荧光PCR检测肝组织miR-214表达水平。结果 H&E和天狼猩红染色结果显示实验组纤维间隔明显。Western blot结果显示实验组α-SMA高表达,同时miR-214agomir干预组α-SMA及Caspase 3表达下调(P<0.05),细胞凋亡及纤维化受到抑制。实时荧光PCR检测结果显示,实验组miR-214在造模6周及8周时表达显著降低(P<0.05)。结论 miR-214高表达抑制细胞凋亡,从而可能起到改善肝纤维化的目的。
Objective To study the effect of miR-214-mediated apoptotic proteinCaspase 3 on liver fibrosis in mice.Methods Six-week-old C57BL/6 mice were randomly divided into two groups:the experimental group was injected with carbon tetrachloride to induce liver fibrosis,and the control group was injected with olive oil.miR-214 agomirwasadministered bytail vein injection.The pathological changes of fibrosis model were evaluated by H&E and Sirius red staining.The expression levelsofα-SMA and Caspase 3 in the liver of the experimental group and the control group were determined by Western blot.Quantitative PCR was used to detect the expression level of miR-214 in liver tissue.Results The H&E and Sirius red staining showed that the experimental group had obvious fiber interval.Western blot results showed that the expression ofα-SMA was high in the experimental group,while the expression ofα-SMA and Caspase-3 was down-regulated in the miR-214 agomir intervention group(P<0.05),and liver fibrosis were inhibited.The results of Quantitative PCR showed that the expression of miR-214 in the experimental group was significantly decreased at 6 and 8 weeks after model establishment(P<0.05).Conclusion UpregulatedmiR-214 inhibits apoptosis,which may improve liver fibrosis.
