摘要
目的采用注射一氧化氮(NO)前体左旋精氨酸(L-Arg),研究实验性牙移动大鼠牙周组织中人类白细胞分化抗原133(CD133)的表达。方法取雄性的6周龄Wistar大鼠40只,随机分为实验组(20只)和对照组(20只),采用50g力拉右上颌第一磨牙向近中移动,建立牙移动动物模型。实验组腹腔注射L-Arg,给药剂量为300mg·kg^(-1)溶解于0.5ml生理盐水,对照组腹腔注射等量生理盐水。实验组和对照组大鼠再分别分为加力1、3、5、7、14d组,每组4只,分别在实验的规定时间处死大鼠并制作以右上颌第一磨牙为中心的、近远中方向的组织切片,进行HE染色观察牙周组织变化、CD133免疫组织化学染色观察CD133在牙周组织中的表达,图象分析并统计数据。结果实验组与对照组牙周组织中均可见CD133不同程度的阳性表达,两组间相同时间点比较,实验组CD133阳性反应灰度积分普遍高于对照组,两组CD133阳性反应灰度积分在加力3、5天时比较差异有统计学意义(P<0.05)。实验组和对照组均可观察到CD133表达阳性的新生血管,实验组在加力3天时CD133阳性的新生血管表达高于对照组,组间比较差异有统计学意义(P<0.05)。实验组CD133在成骨细胞及其前体中表达明显。结论 L-Arg通过上调实验性牙移动牙周组织中CD133的表达,促进CD133阳性细胞参与实验性牙移动的牙周组织改建,从而在促进实验性牙移动牙周组织的血管改建与骨改建方面发挥了作用。
Objective To investigate the expression of CD133 in periodontal tissues of experimental tooth movement rats by injecting NO precursor L-arginine(L-Arg).Methods 6 week old male Wistar rats were randomly divided into experimental group(20 rats)and control group(20 rats).The first molar of the right maxilla was pulled to move mesially with 50 g force to establish an animal model of tooth movement.The experimental group was injected intraperitoneally with L-Arg at a dose of 300 mg·kg^-1 dissolved in 0.5 ml of normal saline,while the control group was injected with the same amount of normal saline.The experimental group and the control group were divided into the 1,3,5,7,and 14-day groups,with 4 rats in each group.The rats were sacrificed at the prescribed time of the experiment,and the tissue sections of the maxillary first molar were made.The changes of periodontal tissues were observed by HE staining,the expression of CD133 in periodontal tissues was observed by CD133 immunohistochemical staining,and the image analysis and statistical data were obtained.Results The positive expression of CD133 in the periodontal tissues of both the experimental group and the control group was observed.Comparing the two groups at the same time point,the gray scale integral of CD133 positive reaction in experimental group was generally higher than that in control group.The intensity of CD133 expression in the experimental group was significantly higher than that in the control group at third day and fifth day(P<0.05).Both the experimental group and the control group were able to observe the expression of CD133 positive new blood vessels.The expression of CD133 positive new blood vessels in the experimental group on the third day was significantly higher than that of the control group(P<0.05).The expression of CD133 in osteoblasts and their precursors in the experimental group was obvious.Conclusion L-Arg promotes the involvement of CD133 positive cells in periodontal tissue remodeling in experimental tooth movement by up-regulating the expression of CD133 in periodontal tissue,thus playing a role in promoting vascular remodeling and bone remodeling in periodontal tissue of tooth movement.
