木芙蓉总黄酮提取工艺及季节变化规律研究 被引量:6

Study on Extraction and Content Variation of Total Flavonoids from Hibiscus mutabilis Leaf

摘要 目的采用响应面法优选木芙蓉叶总黄酮的工艺,并通过测定每月总黄酮了解木芙蓉叶月周期变化规律。方法在单因素的基础上,以乙醇浓度、液料比、超声时间为自变量,木芙蓉叶总黄酮得率为因变量,通过响应面法实验,用Design-Expert 8.0.6软件对实验数据进行分析优化最佳工艺。结果总黄酮最佳提取工艺为乙醇浓度46%,液料比33∶1,超声时间34min。4月份木芙蓉叶总黄酮44.5538mg/g为最高,8月份其次。结论响应面法来优选的提取简便、准确、可靠,可运用此法来提取木芙蓉叶总黄酮。结合总黄酮含量和叶片产量来考虑8月份可以作为木芙蓉叶片最佳采收期。 Objective Response surface methodology was used to optimize the extraction conditions of total flavonoids from Hibiscus mutabilis Leaf.In order to understand periodic variation of total flavonoids in the leaves of Hibiscus was determined the per month.Methods Based on one factor test,the individual and interactive effects of ethanol concentration,material/liquid ratio and extracting time on the yield of flavonoids were studied.Results The experiments showed that the optimal extracting conditions were as follows:ethanol concentration 46%,material/liquid ratio 1:33 and extraction time 34 min.Under these conditions,the highest yield of flavonoids was 44.5538 mg/g in April and high yield was August.Conclusion The optimum extraction method of response surface method is simple,accurate and reliable.This method can be used to extract total flavonoids from the leaves of Hibiscus.Considering the total flavones content and leaf yield,the best harvest time of leaves could be considered in August.
