黄芩苷对2型糖尿病肾病大鼠survivin表达的影响 被引量:8

The effect of baicalin on expression of survivin in kidney of type 2 diabetic rats

摘要 目的研究黄芩苷对糖尿病(DM)大鼠肾组织凋亡相关因子survivin表达的影响,探讨其对肾脏的保护作用。方法健康成年Wistar大鼠30只,随机分为正常对照组(NC组,n=10)、糖尿病模型组(DM组,n=10)和黄芩苷干预组(BAI,n=10)。6周后观察各组血糖、血脂水平;一侧肾脏做组织形态学检查,另一侧肾脏液氮保存后采用荧光定量RT-PCR的方法测survivin mRNA的表达。结果肾脏免疫组化和荧光定量RT-PCR结果均显示,黄芩苷组survivin表达增加,与模型组相比较,差异有显著性差异(P<0.05)。结论黄芩苷可减轻DM大鼠肾脏细胞的凋亡,延缓DN的发展进程,其作用机制可能是通过增加凋亡抑制因子survivin的表达,从而提高肾脏抗凋亡系统的功能。 Objective To investigate the effect of baicalin on apoptosis and the expression of apoptotic regulatory protein survivin in kidney of type 2 experimental diabetic rats.So to ascertain the anti-apoptosis of baicalin in the diabetic rat kidney.Methods Thirty healthy male Wistar rats were divided randomly into three groups:the normal control group(n=10),the diabetes group(n=10)and the baicalin group(n=10).After 6 weeks,all rats were weighted,the fasting blood glucose,triglyceride,cholesterol,high-density lipoprotein,low-density lipoprotein were determined.Reverse transcription-polymerase chain reaction was used for the determination of mRNA for survivin of kidney.Immunohistochemistry was used for the determination of protein expression for survivin of kidney.Results Compared with the normal control group,the mRNA and protein expression of survivin of kidney was significantly lower in the diabetes group and the baicalin group(P<0.01).Compared with the diabetes group,the protein and the mRNA expression of survivin of kidney was significantly higher(P<0.05)in the baicalin group.Conclusion Baicalin can reduce the apoptosis of renal cell in diabetic rats,the mechanism might be baicalin can up-regulate the expressions of inhibitor of apoptosis protein survivin in diabetic rat,then improve the function of the system of anti –apoptosis in the kidney in the diabetic rats.
