摘要
目的观察hsa_circ_0005692在胃癌中的表达及其对胃癌细胞迁移、侵袭和增殖的影响,探讨hsa_circ_0005692在胃癌中的生物学功能。方法选取8例胃癌及相应癌旁组织进行基因芯片筛查,随机筛选10种表达下调的环状RNA(circRNA),并利用31例胃癌组织及相应癌旁组织中应用反转录-聚合酶链反应(RT-PCR)进行验证。通过划痕实验检测细胞的迁移能力,采用Transwell侵袭实验检测细胞的侵袭能力,采用细胞计数试剂盒(CCK-8)法测定胃癌BGC-823细胞增殖。结果在胃癌及癌旁组织中差异表达下调(差异≥1.5倍,P<0.05)的circRNA共761种,RT-PCR进一步验证显示,hsa_circ_0089762、hsa_circ_0036592、hsa_circ_0005692、hsa_circ_0000141 4种circRNA的相对表达量分别为0.431±0.163、0.689±0.110、0.631±0.165、0.633±0.171,差异均有统计学意义(P<0.05)。细胞划痕实验、侵袭实验、增殖实验结果显示,与对照组比较,过表达hsa_circ_0005692胃癌BGC-823细胞的迁移、侵袭和增殖能力显著降低,差异均有统计学意义(P<0.05)。结论hsa_circ_0005692在胃癌中表达下调,且可抑制胃癌细胞的迁移、侵袭和增殖。
Objective To identify the differentially expressed circular RNA (circRNA) in gastric cancer and explore the biological function of hsa_circ_0005692. Methods Eight cases of gastric cancer and their adjacent tissues were selected to detect the differentially expressed circRNA by circRNA microarray analysis. The changes of 10 selected circRNAs were confirmed by reverse transcriptase-polymerase chain reaction (RT-PCR). The migration ability was analyzed by wound healing assay. The invasive ability was analyzed by Transwell assay. The proliferation of the cells was measured by cell counting kit-8 (CCK-8). Results A total of 761 downregulated circRNAs (fold-change,>1.5, P<0.05) were identified in gastric cancer as compared with the adjacent normal gastric tissues. The RT-PCR showed that the expression of hsa_circ_0089762 (0.431±0.163), hsa_circ_0036592 (0.689±0.110), hsa_circ_0005692 (0.631±0.165) and hsa_circ_0000141 (0.633±0.171)(P<0.05, respectively) in gastric cancer was significantly lower than that in adjacent normal tissues. The wound healing assay, Transwell assay and cell viability assay showed that over-expression of hsa_circ_0005692 could suppress the migration, invasion and proliferation of BGC-823 cells as compared with the control group (P<0.05, respectively). Conclusion The expression of hsa_circ_0005692 is down-regulated in gastric cancer, and it can inhibit the migration, invasion and proliferation of gastric cancer. It is expected that hsa_circ_0005692 could be a new therapeutic strategy for gastric cancer patients, and the molecular mechanism of hsa_circ_0005692 in gastric cancer progression merits further investigation.
