电针对食源性肥胖大鼠下丘脑自噬相关蛋白7表达的影响 被引量:10

Effect of Electroacupuncture on Hypothalamic Autophagy-related Protein 7 Expression in Diet-induced Obesity Rats

摘要 【目的】探讨电针对食源性肥胖大鼠下丘脑自噬相关蛋白7(Autophagy-related protein 7,Atg7)表达的影响。【方法】将40只雄性SD大鼠随机分为正常对照组8只和饮食诱导肥胖(Diet-induced obesity,DIO)造模组32只,造模组给予高脂饲料饲养14周制备DIO大鼠肥胖模型,取造模成功的16只大鼠随机分为肥胖模型组和针刺干预组,每组各8只。针刺干预组选取中脘、足三里、天枢、三阴交穴进行治疗,每天1次,每周连续针刺5 d,共针刺干预4周。观察各组大鼠体质量的变化,Western blot法检测大鼠下丘脑Atg7蛋白质的表达。【结果】高脂喂养14周后肥胖模型组和针刺干预组大鼠体质量均明显高于正常对照组,差异有统计学意义(P <0.05)。电针干预4周后,针刺干预组大鼠体质量显著低于肥胖模型组(P <0.05)。肥胖模型组大鼠下丘脑Atg7蛋白质含量明显低于正常对照组(P <0.05),电针干预组大鼠下丘脑Atg7蛋白质含量高于肥胖模型组(P <0.05)。【结论】电针干预可减轻肥胖大鼠体质量,可能与调控下丘脑自噬水平变化有关。 Objective To investigate the effect of electroacupuncture(EA)on the expression of autophagy-related protein 7(Atg7)in the hypothalamus of diet-induced obesity(DIO).Methods Forty male SD rats were randomly divided into normal control group(n=8)and model group(n=32).The rats in the model group were fed by high-fat forage for 14 successive weeks and then the 16 modeled rats were divided into obesity model group(n=8)and EA treatment group(n=8).EA treatment group was given EA on points of Zhongwan(RN12),Zusanli(ST36),Tianshu(ST25)and Sanyinjiao(SP6)once a day,for 5 successive days per week and 4 weeks in total.Body weight was measured in all of the rats,and the expression of Atg7 in the hypothalamus was detected by Western blot method.Results After feeding with high-fat forage for 14 weeks,the body weight of rats in the obesity model group and EA treatment group was obviously higher than that in the normal control group(P<0.05).After EA treatment for 4 weeks,body weight in EA treatment group was obviously lower than that in the obesity model group(P<0.05).Protein expression level of Atg7 in the hypothalamus of the obesity model group was lower than that in the normal control group(P<0.05),and EA treatment can increase the expression Atg7(P<0.05 compared with that in the obesity model group).Conclusion EA treatment could reduce the body weight of DIO rat through upregulating the level of Atg7 protein in rat hypothalamus.
