摘要
首次采用高效逆流色谱技术,以乙酸乙酯:石油醚:乙醇:水(1. 25:0. 2:0. 1:1. 4,V/V/V/V)为溶剂体系,从50 g桑叶乙酸乙酯提取物中分离得到52. 1 mg异槲皮苷和32. 7 mg紫云英苷两种黄酮化合物。经高效液相色谱检测,其纯度分别为96. 61%和97. 78%。应用高效液相色谱和电喷雾质谱联用技术,通过与标准化合物的液相色谱保留时间、紫外光谱,分子离子峰及中性碎片的丢失等进行对比,对分离得到的两种化合物的进行结构鉴定。
A total of 52. 1 mg isoquercitrin and 32. 7 mg astragalin were successfully isolated and purified from 50 g ethyl acetate extract of Morus alba for the first time by high performance currentcounter chromatography ( HPCCC) using the solvent system ethyl acetate: petroleum benzin : ethanol: water at the volumn ratio of 1. 25 : 0. 2 : 0. 1 : 1.4( V/V/V/V), and the purities of two compounds determined by HPLC were 96. 61 % and 97. 78%, respectively. The structures of the two compounds were identified by congruent retention times, UV spectra, the data of the liquid chromatography-electrospray ionization-mass spectrometry ( LC-ESI-MS) in the negative ion mode with those of the authentic standards and literature reports.
