摘要
将荧光内滤效应与滚环扩增放大技术相结合,构建了黏蛋白检测方法。二氧化锰纳米片可淬灭MoS2量子点荧光,加入谷胱甘肽溶解MnO2荧光恢复。通过滚环扩增产生大量的核酸酶可催化谷胱甘肽氧化,使其无法与MnO2反应而抑制荧光。适配体与黏蛋白作用后将阻碍滚环扩增反应进行,促进荧光恢复。在0. 5~80 nmol/L范围内,黏蛋白浓度与荧光强度呈良好的线性关系,检测限为15 pmol/L。该方法对黏蛋白检测具有良好的选择性及重现性。
A novel highly sensitive and selective method for detecting mucin was constructed by combining fluorescence inner filtration effect and rolling circle amplification technology. The fluorescence of molybdenum disulfide quantum dots( MoS2 QDs) could be quenched by manganese dioxide nanosheets( MnO2 NSs). Glutathione was added to dissolve manganese dioxide and restore the fluorescence. Through rolling circle amplification,a large number of nuclease could be produced and catalyze the oxidation of glutathione. Thus,it could not react with MnO2 NSs and the fluorescence was inhibited. The interaction of aptamer and mucin would block the reaction of rolling circle amplification and promote the recovery of fluorescence. A good linear relationship was obtained between the concentration of mucin( 0. 5-80 nmol/L) and fluorescence intensity. The detection limit was estimated to be 15 pmol/L. The method has good selectivity and reproducibility for the detection of mucin.
