慢性荨麻疹患者外周血TLR信号分子的变化及临床意义 被引量:6

Changes of TLR signaling molecule in peripheral blood of patients with chronic urticaria and its clinical significance

摘要 目的研究慢性荨麻疹患者外周血Toll样受体(TLR)信号分子的变化及临床意义。方法选择2014年6月至2018年3月期间在本院诊断为自身免疫性慢性荨麻疹(AIU)的患者作为研究的AIU组,另选取同期在本院体检的健康者作为对照组。采集肘静脉血并测定TLRs信号分子TLR2、TLR4、TLR9的mRNA相对表达量以及Th1、Th2、Th17、Treg细胞因子的含量。采用两独立样本t检验分析。结果 AIU组患者外周血中TLR2、TLR4、TLR9的mRNA相对表达量以及血清中白细胞介素-4(IL-4)、IL-5、IL-17、IL-10、转化生长因子-β1(TGF-β1)、IL-35的含量均显著高于对照组(P=0. 000),血清中干扰素-γ(IFN-γ)、IL-2的含量显著低于对照组(P=0. 000); Pearson检验显示,AIU组患者外周血中TLR2、TLR4、TLR9的相对mRNA表达量与血清中IL-4、IL-5、IL-17、IL-10、TGF-β1、IL-35的含量呈正相关,与血清中IFN-γ、IL-2的含量呈负相关。结论 AIU患者外周血TLR信号分子TLR2、TLR4、TLR9的高表达能够造成病程中Th1/Th2/Th17/Treg免疫应答发生紊乱。 Objective To study the mRNA expression of Toll like receptor (TLR) signaling molecule in peripheral blood of patients with chronic urticaria. Methods The patients who were diagnosed with autoimmune chronic urticaria (AIU) during June 2014 - March 2018 were selected as the AIU group, and the healthy persons receiving medical examination in the same period were selected as the control group. The venous blood was collected to detect the mRNA expression of TLRs, TLR2, TLR4 and TLR9 and the contents of Th1, Th2, Th17 and Treg cytokines. Results The mRNA expression of TLR2, TLR4 and TLR9 in the peripheral blood and the content of interleukin-4 (IL-4), interleukin-5 (IL-5), interleukin-17 (IL-17), interleukin-10 (IL-10), transforming growth factor -β1 (TGF-β1), and interleukin-35 (IL-35) in AIU group were significantly higher than those in the control group (P=0.000), while the content of interferon-γ(IFN-γ) and interleukin-2 (IL-2) in AIU group 0were significantly lower than those in the control group (P=0.000). The mRNA expression of TLR2, TLR4 and TLR9 were positively correlated with the serum contents of IL-4, IL-5, IL-17, IL-10, TGF-β1, IL-35 and negatively correlated with the serum contents of IFN-γ, IL-2. Conclusions The high mRNA expression of TLR2, TLR4 and TLR9 in peripheral blood of AIU patients can cause disorder of Th1/Th2/Th17/Treg immune response in the course of disease.
