摘要
目的:通过测定紫外线照射人真皮微血管内皮细胞(HDMEC)建立的光损伤模型中清除自由基的酶SOD、CAT和细胞间黏附分子ICAM-1的含量,探讨中药汤剂桃红四物汤对UVA诱导的人真皮微血管内皮细胞光损伤后自由基形成和细胞间黏附分子的影响,进而探讨桃红四物汤对光损伤模型中人真皮微血管内皮细胞的保护作用,并从细胞分子层次阐明桃红四物汤拮抗光损伤的作用机理,为皮肤光损伤应用桃红四物汤来治疗提供了进一步深入研究的依据。方法:建造HDMEC细胞的培养与传代模型,通过ELISA方法检测抗氧化损伤标志物超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、细胞间黏附分子(ICAM-1)来研究桃红四物汤在离体人类细胞层次上对于治疗光老化的作用。结果:UVA照射可使HDMEC细胞分泌CAT、SOD降低并提高ICAM-1表达水平。且桃红四物汤高、中、低剂量含药血清均能够促进HDMEC细胞分泌CAT、SOD升高并抑制ICAM-1表达水平,高剂量组效果最好。
Objective: This research through the detection of free radicals in light damage model, enzyme SOD, CAT and the content of Intercellular adhesion molecule ICAM - 1 , explored Taohong Siwu Decoction on free radicals and intercellular adhesion molecules of the human dermal icrovascular endothelial cells induced by UVA light damage and discussed the protection of Taohong Siwu Decoction on human dermal microvascular endothelial cells caused by ultraviolet irradiation damage, providing the experimental basis for using Taohong Siwu Decoction in the treatment of light damage. Method: The culture and transmission model of HDMEC cells was set up. The ELISA method was used to detect antioxidant injury markers superoxide dismutase ( SOD), catalase ( CAT) and intercellular adhesion molecule (ICAM - 1 ) to study the effect of Taohong Siwu Decoction on light aging in vitro human cell level. Conclusion: The UVA irradiation can reduce HDMEC cell secretion of CAT, SOD and raise the level of expression of ICAM - 1. Taohong Siwu Decoction high, medium and low doses medicated serum can promote the HDMEC cell secretion of CAT, increase SOD and inhibit ICAM - 1 expression and the high dose group is the best.
