益气活血方防治糖尿病肾病足细胞损伤的实验研究

Experimental Study on Protective Effect of YiqiHuoxue Recipe on Podocyte Injury of Diabetic Nephrology

目的:研究益气活血方对链脲菌素(Streptozocin,STZ)诱导的糖尿病肾病(Diabetic nephrology,DN)足细胞损伤的保护作用。方法:8周龄雄性SD(Sprague-Dawley)大鼠采用STZ造模成功后随机分为模型组和益气活血方组,同时设立正常对照组,每组8只,益气活血方组给予黄芪颗粒0.8 g/(kg·d)联合三七颗粒0.4 g/(kg·d)生理盐水溶液灌胃,对照组和模型组给予等量生理盐水溶液灌胃,连续12周。12周末,收集24 h尿液检测尿微量白蛋白浓度,光镜观察肾脏病理改变,电镜观察足细胞超微结构变化,并采用免疫组织化学方法检测肾组织中nephrin表达。结果:与模型组相比,益气活血方组可明显减轻DN大鼠尿蛋白排泄(P<0.05)和肾脏病理改变,并减轻足细胞足突融合及改善足细胞裂孔隔膜蛋白nephrin表达。结论:益气活血方能减轻DN足细胞损伤和蛋白尿,改善肾脏病理损伤,从而发挥肾脏保护作用。

Objective: To study the protective effect of YiqiHuoxue Recipe on podocyte injury in diabetic rats. Methods :Diabetes was induced with streptozocin. After that, diabetic rats were randomly divided into DN group and YiqiHuoxue Recipe group, together with the normal control group, 8 rats in each group. YiqiHuoxue group rats were treated with Huangqi Granule [0. 8 g/( kg·d)] and Sanqi Granule [0. 4 g/( kg·d)], while the other two groups were treated with isometric volume of saline. All rats were gavaged for 12 weeks. After treatment, 24 hours urine was gathered for the detection of urinary albumin and kidneys were collected for renal histopathology study. Results: Compared with DN group, the renal histopathology was obviously improved and the expression of nephrin was upregulated in YiqiHuoxue Recipe group. Conclusion: YiqiHuoxue Recipe can relieve podocyte injury and albuminuria and improve kidney histopathological injury, resulting in renal protective effect in diabetic rats.
