摘要
布鲁氏菌病是由布鲁氏菌引起的以流产和发热为主要特征的人兽共患病,严重威胁着人和多种动物的生命健康,导致巨大经济损失及严重危害公共安全。实验室前期利用噬菌体展示技术建立布鲁氏菌16M株基因组文库,以期从该文库中筛选具有良好抗原性的诊断制剂。为检测构建文库的随机性及多样性,本研究利用Touch-down PCR对构建的噬菌体展示布鲁氏菌基因组文库进行评价。结果表明,构建文库具有较好随机性及多样性,为后续试验奠定基础。
Brucellosis is a zoonosis caused by Brucella spp., which is characterized by abortion and fever. It seriously threatens the life and health of human beings and many animals, resulting in huge economic losses and serious public health hazards. The Brucella melitensis 16M strain genomic library has been established by phage display technique to screen diagnostic agtigens with good antigenicity. To detect the randomness and diversity of the library, Touch-down PCR was used to evaluate the phage-displayed library. The results show that the library has good randomicity and diversity, and lays the foundation for subsequent experiments.
