不同品种赤小豆子叶SOD提取技术优化及活性研究 被引量:4

摘要 以京农红8号赤小豆子叶为材料,进行单因素和正交试验,优化出超声波法提取赤小豆子叶超氧化物歧化酶(SOD)的最佳条件,并分析比较不同品种赤小豆子叶中SOD的活力。结果表明,赤小豆子叶SOD最佳提取工艺条件为:液固比15∶1(m L/g),超声提取功率140 W,磷酸缓冲液pH值7.0,水浴浸提温度40℃;对赤小豆子叶SOD提取率影响的大小顺序为超声功率>磷酸缓冲液pH值>液固比>水浴温度。对比8个品种的赤小豆子叶SOD活力可以得出,不同品种赤小豆子叶SOD含量有显著差异,小丰7号SOD活力最高,SOD活力值为11.17 U/g;而吉红2号SOD活力最低,活力值仅为8.97 U/g。 This paper used Jingnonghong 8 red bean cotyledons as experimental material,through single-factor experiment and orthogonal test to find the optimum conditions in the ultrasonic extraction of superoxide dismutase(SOD). Meanwhile, the activities of SOD in different varieties of red bean were compared and analyzed. The results showed that the optimum extraction conditions of SOD from red bean cotyledons were as follows: liquid-solid ratio 15∶1(mL/g), ultrasonic extraction power 140 W, phosphate buffer pH 7.0, and water bath extraction temperature 40 ℃. The influence of four factors on SOD extraction rate of red bean cotyledons was ultrasound power>pH value of phosphate buffer>liquid-solid ratio>water bath temperature. Comparing the SOD activity of cotyledons of eight red bean varieties, the results showed that different varieties of red beans were significantly different. The SOD activity of Xiaofeng 7 was the highest, the SOD activity value was 11.17 U/g, while Jihong 2 was the lowest, the SOD activity value was only 8.97 U/g.
