单硝酸异山梨酯缓释片对慢性高原病模型大鼠多脏器损伤的改善作用

Ameliorative effect of Isosorbide Mononitrate Sustained Release Tablets on multiple organ injury in rats with chronic mountain sickness

摘要 目的探讨单硝酸异山梨酯缓释片(IMSRT)对慢性高原病(CMS)大鼠多脏器损伤的改善作用。方法将60只SD大鼠随机抽取50只,放入模拟海拔5 000 m的高原环境中饲养30 d,进行CMS造模,剩余10只在平原环境中饲养30 d;高原环境大鼠随机分为模型组(蒸馏水4 mL/kg体质量)、硝苯地平组(2.7 mg/kg体质量)、单硝酸异山梨酯缓释片高、中、低剂量组(3.24、1.62、0.81 mg/kg体质量),平原环境大鼠为正常组(蒸馏水4mL/kg体质量),灌胃15 d后,测定各组大鼠的肺动脉压力(PAP),观察各组大脑、肾脏、肺病理学改变,测定肾、肺组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)含量。结果与正常组大鼠比较,模型组、硝苯地平、IMSRT各剂量组大鼠PAP升高,大鼠肺、肾脏组织中MDA水平升高,SOD和GSH-Px活性降低,差异有统计学意义(P<0.05)。与模型组大鼠比较,IMSRT各剂量组大鼠PAP降低,大鼠肺、肾脏组织中MDA水平降低,SOD和GSH-Px活性升高,差异有统计学意义(P<0.05)。结论 IMSRT对CMS大鼠的多脏器损伤具有一定改善作用。 Objective To investigate the ameliorative effect of Isosorbide Mononitrate Sustained Release Tablets(IMSRT) on multiple organ injury in rats with chronic mountain sickness(CMS). Methods 50 SD rats were fed for 30 days at simulated altitude of 5 000 M. 10 SD rats were fed for 30 days in plain environment. The rats in plateau environment were randomly divided into model group(4 mL/kg body weight of distilled water), nifedipine group(2.7 mg/kg body weight), high, medium and low dose of isosorbide mononitrate sustained-release tablets groups(3.24, 1.62 and 0.81 mg/kg body weight). The rats in plain environment were the normal group(4 mL/kg body weight of distilled water). After 15 days, the pulmonary artery pressure(PAP) was measured. The pathological changes of brain, kidney and lung were observed. The contents of superoxide dismutase(SOD), malondialdehyde(MDA) and glutathione peroxidase(GSH-Px) in kidney and lung were measured. Results Compared with the normal group, PAP and MDA level in lung and kidney tissues of rats was increased significantly(P<0.05), while the activity of SOD and gsh-px was decreased significantly(P<0.05), in the model group, nifedipine and IMSRT groups. Compared with the model group, PAP and MDA level in lung and kidney tissues of rats was decreased significantly(P<0.05), while the activity of SOD and gsh-px was increased significantly(P<0.05), in each dose group of IMSRT. Conclusion IMSRT could improve multiple organ injury in CMS rats.
