摘要
目的探讨肾透明细胞癌组织CDK9的表达水平,分析其与肾透明细胞癌临床特征的关系,研究CDK9对肾细胞癌细胞迁移和侵袭能力的影响。方法选取2013年1月至2016年12月在河南科技大学第一附属医院接受手术切除治疗的肾透明细胞癌患者组织标本102例和癌旁组织标本30例作为研究对象,应用实时荧光定量PCR和Western blot分别检测CDK9的表达水平,收集102例患者性别、年龄、病理分级等临床病理资料,并分析其与CDK9表达的关系。体外应用肾细胞癌细胞(ACHN、786-O和OSRC-2)研究CDK9对肾细胞癌细胞迁移和侵袭能力的影响。结果与癌旁组织比较,CDK9 mRNA和蛋白在肾透明细胞癌组织中表达显著上调(P<0.01)。CDK9 mRNA表达水平以中位数为界分为低表达组和高表达组,两组患者的病理T分期、TNM分期和远隔转移比较,差异有统计学意义(P<0.05)。肿瘤细胞系中CDK9表达显著高于人正常肾小管上皮细胞HK-2,其中786-O及ACHN细胞中CDK9表达显著升高(P均<0.01)。体外沉默CDK9显著抑制786-O及ACHN的细胞增殖、迁移和侵袭(P<0.01)。结论 CDK9表达与肾透明细胞癌的恶性特征、癌细胞增殖、迁移和侵袭能力密切相关,有望成为肾透明细胞癌的潜在治疗靶点。
Objective To investigate the expression of CDK9 in renal cell carcinoma and analyze its correlations with the clinical characteristics of renal cell carcinoma.To study the effect of CDK9 on the migration and invasion of renal cell carcinoma cell lines.Methods Samples from 102 cases of renal cell carcinoma patients who received surgical resection and 30 cases of adjacent tissues were collected at the First Affiliated Hospital of Henan University of Science and Technology from January 2013 to December 2016.The real-time quantitative PCR and Western blot were used to detect the expression of CDK9,and the clinicopathological data of 102 patients including sex,age and pathological grade were collected and analyzedfor their potential associations with CDK9 expression.The effects of CDK9 on the migration and invasion of renal cell carcinoma cells were studied using renal cell carcinoma cells(ACHN,786-O and OSRC-2)in vitro.Results CDK9 mRNA and protein were significantly up-regulated in renal cell carcinoma tissues compared with adjacent tissues(P<0.01).The expression level of CDK9 mRNA was divided into low-expression group and high-expression group by median.The pathological T-stage,TNM stage and distant metastasis of the two groups were statistically significant(P<0.05).The expression of CDK9 in tumor cell lines was significantly higher than that in normal renal tubular epithelial cells(HK-2),and the expression of CDK9 in 786-O and ACHN cells was significantly increased(P<0.01).In vitro silencing of CDK9 significantly inhibited cell proliferation,migration and invasion of 786-O and ACHN.Conclusion The expression of CDK9 is closely related to the aggressive features of renal cell carcinoma and the proliferation,migration and invasion of cancer cells,which is expected to be a potential therapeutic target for renal cell carcinoma.
