摘要 目的探讨羟基磷灰石(HA)涂层及成骨多肽(BFP)接枝双重改性方法对聚醚醚酮(PEEK)表面生物活性的影响。方法将3D打印的PEEK(作为PEEK组)浸泡于聚多巴胺(PDA)溶液,24 h后取出,作为PDA-PEEK组;转移至1.5倍模拟体液(SBF)中7 d,完成HA涂层,作为HA-PEEK组;HA-PEEK试样浸泡于PDA溶液24 h,作为PDA/HA-PEEK组;再转移至BFP多肽溶液,完成BFP接枝,作为HA-BFP-PEEK组。分别测定各组材料的表面接触角;各组材料分别与成骨细胞MG63共培养,通过CCK-8、碱性磷酸酶(ALP)活性、茜素红染色实验评估细胞的附着、增殖能力及成骨活性。结果与PEEK组相比,改性后各组接触角减小,细胞附着和增殖能力均增强,ALP活性提高,钙结节形成增多(P <0.05),且HA-BFP-PEEK组明显优于其他各组(P <0.05)。结论 PEEK经HA涂层及BFP接枝双重改性后,生物活性显著提高,有望为PEEK提供一种新的表面改性方式。 Objective To explore the effect of double modification methods of hydroxyapatite(HA)coating and osteogenic polypeptide(BFP)grafting on the surface biological activity of polyether ether ketone(PEEK).Methods 3D printed PEEK(“PEEK group”)was immersed in polydopamine(PDA)solution for 24 hours and then removed(“PDA-PEEK group”);after it was transferred to 1.5 times simulated body fluid(SBF)and maintained for seven days,HA coating was completed(“HA-PEEK group”);HA-PEEK samples were immersed in PDA solution for 24 hours(“PDA/HA-PEEK group”),and then transferred to BFP solution to complete BFP grafting(“HA-BFP-PEEK group”).The surface contact angles of each group were measured,and the materials were co-cultured with osteoblasts MG63.The adhesion,proliferation and osteogenic activity of the cells were evaluated by CCK-8,alkaline phosphatase(ALP)activity and alizarin red staining.Results Compared with the PEEK group,the contact angle decreased and the cell attachment,proliferation,ALP activity and calcium nodule formation increased(P<0.05),and the HA-BFP-PEEK group was significantly better than other groups(P<0.05).Conclusion The bioactivity of PEEK modified by HA coating and BFP grafting is significantly improved,which is expected to provide a new surface modification method for PEEK.
