养血清脑颗粒治疗脑卒中神经元损害的分子机制 被引量:9

摘要 目的探究养血清脑颗粒治疗脑卒中大鼠神经元损害的机制。方法 60只SD大鼠随机分为假手术组、模型组、实验1组和实验2组,各15只。假手术组不插拴线,其余3组采用线栓法建立缺血性脑卒中大鼠模型。在造模手术后3 d,假手术组与模型组均采用生理盐水1 ml/(100 g·d)灌胃,实验1组以4.0 g/(100 kg·d)的养血清脑颗粒灌胃,实验2组为8.0g/(100 kg·d),均1次/d,连续7 d。在灌胃后1、5、7 d,行神经症状缺损评分;灌胃后7 d,测定血清神经元特异性烯醇化酶(NSE)、同型半胱氨酸(Hcy)、神经生长因子(NGF)、脑源性神经生长因子(BDNF)、神经肽Y(NPY)、强啡肽(Dyn)含量。结果假手术组神经症状缺损评分均为0;在干预后各时间点,模型组神经症状缺损评分最高,其次为实验1组,实验2组最低(P <0.05),且随着干预时间的延长,模型组神经症状缺损评分无显著变化(P> 0.05),而实验1组和实验2组评分逐渐降低,表明神经缺损程度逐渐减轻。灌胃后7 d,4组血清NSE、Hcy、NPY、Dyn对比,均为模型组最高,然后依次为实验1组、实验2组、假手术组(P <0.05);血清NGF、BDNF则为模型组最低,然后依次为实验1组、实验2组、假手术组(P <0.05)。结论养血清脑颗粒能够缓解脑卒中后神经功能缺损症状,调节血清中神经因子的含量,从而改善脑卒中后对神经元的损害,且存在时间-剂量依赖性,值得在临床上应用。 Objective To explore the mechanism of Yangxue Qingnao granule in the treatment of neuron damage in stroke rats.Methods A total of 60 SD rats were randomly divided into a sham-operated group,a model group,experiment group 1 and experiment group 2(n=15,respectively).The sham-operated group did not use embolization,while the other three groups used embolization method to establish ischemic stroke rat models.Three days after the operation,the sham-operated group and the model group were given 1 ml/(100 g·d)saline intragastric administration,while experiment group 1 was given 4.0 g/(100 kg·d)Yangxue Qingnao granules,and experiment group 2 was given 8.0 g/(100 kg·d),once a day for seven days.Neurological symptoms were scored one,five and seven days after gastric administration.Serum levels of neuron-specific enolase(NSE),homocysteine(Hcy),nerve growth factor(NGF),brain-derived nerve growth factor(BDNF),neuropeptide Y(NPY),dynorphin(Dyn)were measured seven days after gastric administration.Results The neurological deficit score of the sham-operated group was 0.At each time point after intervention,the neurological deficit score of the model group was the highest,followed by experiment group 1 and the experiment group 2(P<0.05).With the prolongation of the intervention time,the neurological deficit score of the model group had no significant change(P>0.05).The scores of experiment group 1 and experiment group 2 decreased gradually,indicating the course of neurological deficit.Gradually the degree decreases.Seven days after intragastric administration,NSE,Hcy,NPY and Dyn in the four groups were the highest in the model group,followed by experiment group 1,experiment group 2 and sham-operated group(P<0.05),while serum NGF and BDNF were the lowest in the model group,followed by experiment group 1,experiment group 2 and sham-operated group(P<0.05).Conclusion Yangxue Qingnao granule can relieve the symptoms of neurological deficit after stroke,regulate the content of neurokines in serum,and improve the damage of neurons after stroke.It has a time-dose dependence and is worthy of clinical application.
