金纳米粒子用于小鼠乳腺癌光声成像研究

摘要 目的构建可用于光声(Photoacoustic Imaging,PA)成像的金纳米粒子,探索其成像肿瘤的可行性及PA成像肿瘤的特点。方法我们以牛血清白蛋白为骨架合成金纳米星。本研究选择测多种方式对其进行表征。4T1乳腺癌荷瘤鼠经尾静脉给药后,于不同时间点行肿瘤PA成像。结果纳米粒子呈星状结构,大小比较均匀,平均粒径为86 nm,表面等离子体共振(Surface Plasmon Resonance,SPR)峰位于775 nm近红外区。MTT实验未见明显生物学毒性。PA成像结果显示给药后肿瘤组织明显强化。结论金纳米星的合成方法简单、可重复性好,展现出良好的生物相容性、光声转化效应。我们证实了一种的新型的可用于肿瘤PA成像的纳米对比剂。 Objective To construct gold nanoparticles for photoacoustic imaging (PA) and explore the characteristics of PA imaging. Methods Bovine serum albumin-capped Au nanoparticles were synthesized from bovine serum albumin. Au nanoparticles were characterized by various methods. PA imaging of 4T1 breast cancer was performed at different time points after intravenous administration. Results The nanoparticles were in star-like structure and uniform size. The nanoparticles had an average diameter of 85 nm and the surface plasmon resonance (SPR) peak was at 770 nm. MTT test and pathological results showed no obvious biological toxicity. PA imaging results showed that the tumor tissue was significantly enhanced after administration. Conclusion The Au nanoparticles exhibites good biocompatibility and excellent photoacoustic imaging effects of tumor.
