复方三草滴丸的急性毒性试验

摘要 目的实验考察复方三草滴丸的急性毒性,评价滴丸剂型的安全性。方法预实验:小白鼠禁食16h,不禁水,后随机分两组,每组10只,雌雄各5只。第一组按96g·kg^(-1)剂量一次性灌胃给药。第二组按192g·kg^(-1)给药,24h后,观察小鼠死亡数量。正式实验:20只小白鼠,随机分为4组:超高剂量组(464g·kg^(-1))、高剂量组(215g·kg^(-1))、中剂量组(100g·kg^(-1))、低剂量组(46.4g·kg^(-1)),雌鼠3只雄鼠2只。结果预实验96g·kg^(-1)组死2只,192g·kg^(-1)组死5只。正式实验超高剂量组(464g·kg^(-1))死5只,高剂量组(215g·kg^(-1))死4只、中剂量组(100g·kg^(-1))死1只。结论复方三草滴丸安全性大,LD_(50)为147g·kg^(-1),可信限区间:95.1~227g·kg^(-1)。 OBJECTIVE To study the acute toxicity of compound Three Herbs Droplet and evaluate the safety of Droplet.METHODS Pre-experimental:the mice were fasted for 16 hours,but they were not allowed to water.After the animals were randomly divided into 2 groups,10 in each group,5 males and 5 females.The first group was given bygavage at a dose of 96 g·kg^-1.The second group was treated with 192 g·kg^-1.After 24 hours,the number of mice died was observed.formal experiment:using 20 mice,the animals are randomly divided into 4 groups of ultra-high dose group(464 g·kg^-1),high dose group(215 g·kg^-1),medium dose group(100 g·kg^-1) In the low-dose group(46.4 g·kg^-1),3 females and 2 males mice.RESULTS Two mice died in the pre-experimental 96 g·kg^-1 dose group,and five died in the 192 g·kg^-1 dose group.Five high-dose groups(464 g·kg^-1) died in the formal experiment,four in the high-dose group(215 g·kg^-1),one in the middle-dose group(100 g·kg^-1).CONCLUSION The compound Three Herbs Droplet is safer with a LD50 of 147 g·kg^-1 and a confidence interval of 95.1-227 g·kg^-1.
