摘要
目的建立川芎中阿魏酸的高效液相测定方法,通过比较不同贮藏时间的川芎中阿魏酸的含量变化来综合评估川芎的质量。方法采用Waters 2695高效液相色谱系统,Diamonsil plus C_(18)-A色谱柱(250mm×4.6mm,5μm),流动相为乙腈-1%冰醋酸(17∶83)等度洗脱,流速1.0mL·min^(-1),柱温30℃,检测波长321nm。结果在0.57μg~71.25μg浓度范围内,阿魏酸呈良好的线性关系;平均回收率为99.0%。测得贮藏时间为4年、3年、2年的川芎中阿魏酸的含量降解率分别为38.4%、7.95%和5.46%。结论川芎药材贮藏时间越长,阿魏酸含量降低越多,且从第四年开始,含量开始急剧下降。
OBJECTIVE To establish a method for the deteminationo of ferulic acid in Ligusticum chuanxiong Hort which has different growth years to evaluate the quality Ligusticum chuanxiong Hort.METHODS An Waters 2695 series HPLC-PDA system was used.A Diamonsil plus C18(250 mm×4.6 mm,5μm) was used as the analytical column.The mobile phase consisted of acetonitrile and 1%glacial acetic acid(17∶83) with flow rate of 1.0 mL·min^-1.The eluates were monitored with uv detector at 321 nm.RESULTS The content of ferulic acid showed a good linear relationship within the concentration range of 0.57~71.25μg,The average recovery was 99.0%.The degradation rate of of ferulic acid in Ligusticum chuanxiong Hort which storaged for 4 years,3 years,2 years were 38.4%、7.95% and 5.46% respectively.CONCLUSION The contents of ferulic acid clearly decreased with time,and at the storage time of four years,the content happened an sharply decline.
